三棱黄酮抗HeLa 宫颈癌:降低分裂期细胞比率诱导细胞凋亡

摘要:

目的:研究三棱的总黄酮(Rhizoma sparganii flavonoids,RSF)对HeLa 宫颈癌细胞的毒理作用及其作用机制。方法:分别检测用含有质量浓度11.7~750μg/mL RSF 的DMEM 培养基培养时,组间HeLa 细胞的增殖活性差异,通过免疫荧光技术、流式细胞术和核型分析方法分析RSF 诱导HeLa 细胞毒理的机制。结果:RSF 含量高于188μg/mL 的DMEM培养基可呈剂量性抑制HeLa 细胞的增殖活性(P < 0.01)、增加细胞凋亡小体的比率(P <0.01);与1% 甲醇对照组相比,RSF-375(RSF 含量为375μg/mL)与RSF-750(RSF 含量为750μg/mL)药物组HeLa 细胞的分裂(M)期细胞比率显著下降(P < 0.01),细胞间的接触生长状态消失,体积增大;在本次实验中,RSF-375 与RSF-750 药物组HeLa 细胞微管(a-tublin)细胞骨架形态的特异性免疫结果与对照组相比,细胞形态呈显著不规则状改 变,微丝突触增多,表现为较高的迁移运动趋势。结论:RSF 是三棱抗HeLa 宫颈癌的有效成分,在本实验中,RSF 可以通过干扰细胞有丝分裂相关的机制显著抑制HeLa 细胞的增殖活性(P < 0.01)并诱导细胞凋亡(P < 0.01)。

Abstract:

Objective: To explore the toxicity effect and mechanisms of Rhizoma sparganii flavonoids (RSF) on HeLa cells. Methods: The effect of RSF at a concentration range of 11.7 to 750μg/mL on the proliferation of HeLa cells was evaluated. The toxic mechanisms were elucidated by immunofluorescence, flow cytometry and karyotype analysis. Results: RSF at a concentration higher than 188μg/mL could significantly (P < 0.01) inhibit the proliferation of HeLa cells in a dose-dependent manner, and increase the proportion of apoptotic bodies (P < 0.01). Compared with the control group, the RSF treatment groups at doses of 375μg/mL (RSF-375) and 750μg/mL (RSF-750) revealed a significant decrease of HeLa cells number at M stage (P<0.01), disappearance of intracellular interaction and an increase of cell volume. In addition, RSF-375 and RSF-750 also showed irregular morphological changes, an increase of synaptics and higher moving trend of microfilament, which were observed in monitoring the specific immnuofluorescence of microtubule protein. Conclusion: RSF is a group of bioactive components against HeLa cervical cancer cells, with the physiological functions of significantly inhibiting proliferation and inducing apoptosis by interfering the mitosis of HeLa cells.
