Abstract: In this research, an analytical method for the determination of promethazine residues in livestock meat and viscera by high performance liquid chromatography-tandem mass spectrometry (HPLC-MS/MS) was established. Samples were extracted with formic acid-acetonitrile, and then the extract was purified on an HLB ion exchange extraction column before being quantitatively analyzed by using multi-reaction monitoring (MRM) mode. The results showed that the method had a good linear relationship in the concentration range of 0.5–50 ng/mL with a correlation coefficient of greater than 0.999. The limit of detection (LOD) and limit of quantitation (LOQ) were 0.1 and 0.3 μg/kg, respectively. The recoveries from spiked samples were 85.7%–102.0%, and the relative standard deviations for precision were 0.9%–7.4% (n = 6). This method is simple and rapid, and can be used for the determination of promethazine in animal-derived foods.
