Immunoregulatory Function of Verbascose on Macrophage Cell Line RAW264.7

Abstract: Objective: To investigate the immunoregulatory function of verbascose on macrophage cell line RAW264.7. Methods: RAW264.7 cells were cultured in a 96-well plate with different concentrations of verbascose (25, 50, 100, 200 μg/mL and 400 μg/mL), and the role of verbascose was evaluated based on the proliferative activity of cultured splenocytes as determined by MTT assay, nitric oxide (NO) production and cytokines (IL-6, IL-1β, IFN-α and IFN-γ) expression levels, neutral red phagocytosis. In the present study, lipopolysaccharide (LPS, 10 μg/mL) was used as a positive control, and treatment without verbascose or LPS was used as a negative control. Results: Verbascose significantly improved the proliferation, phagocytosis, NO level and cytokines expression levels when RAW264.7 cells were cultured for 24 h in the studied concentration range. Verbascose at a dose of 200 μg/mL showed the highest effect. Conclusion: Verbascose has an immunomodulatory effect on macrophage cell line RAW264.7 as a potential immunomodulatory enhancer.
