Study on separation and purification technology of total flavonoids from cinnamon with the method of macroreticular resins

Abstract: The separation and purification of cinnamon total flavonoids were studied with the method of macroreticular resins. The major factors which had effects on static and dynamic adsorption and desorption of macroreticular resins were investigated. The optimal separation and purification technology conditions of cinnamon total flavonoids were determined. The results showed that HPD-500 was the ideal macroreticular resin to separation and purification,the optimal technology conditions of static adsorption and desorption to cinnamon total flavonoids using HPD-500 macroreticular resin were as follows: adsorption equilibrium time 6 h, desorption equilibrium time 2 h, sample solution mass concentration 0.8 mg/mL, pH value 6.0 and taking volume fraction 80% ethanol as eluent; the optimal technology conditions of dynamic adsorption and desorption to cinnamon total flavonoids using HPD-500 macroreticular resin were as follows: sample solution mass concentration 1.2 mg/mL, sample solution flow rate 1.5 mL/min, eluent flux 2.0 mL/min and it was eluted completely when the eluent volume was 45 mL. The content of total flavonoids obtained under the optimal conditions reached 91.81%.
