Hepatoprotective Effect of Pleurotus ostreatus Polysaccharides on Carbon Tetrachloride-Induced Liver Injury in Male Kunming Mice

Abstract: In this experiment, Pleurotus Ostreatus polysaccharide (POP) was extracted by hot water and the protective effects of POP on carbon tetrachloride-induced liver injury in mice were studied. 48 male mice were randomly divided into 6 groups and administered with drug continuously for 28 days according to the dose. On the 22nd and 23rd day, the mice except the blank control group induced liver injury by intraperitoneal injection of CCl4. All of the mice were sacrificed on the 29th day, the body weight was weighed, the blood was collected and the organs were fetched. The contents of alanine aminotransferase (ALT), aspartate aminotransferase (AST), alkaline phosphatase (ALP), total cholesterol (TC), triglyceride (TG), low-density lipoprotein cholesterol (LDL-C) and high-density lipoprotein cholesterol (HDL-C) in serum were measured by commercial kit. Then the liver index was calculated and liver tissue homogenate was prepared. The content of malondialdehyde (MDA), total antioxidant capacity (T-AOC) and enzymatic activities of total superoxide dismutase (T-SOD), catalase (CAT) and glutathione peroxidase (GSH-PX) were measured by commercial kit. Compared with model group, the body weight of mice administered with POP decreased slower and the swelling degree of liver decreased significantly, the level of ALT, AST, ALP, TC, TG and LDL-C of serum from mice administered with POP decreased significantly, the content of MDA of the mice decreased significantly and the enzymatic activities of T-SOD, T-AOC, CAT and GSH-PX enhanced markedly. Therefore, POP has obvious protective effect on liver injury induced by carbon tetrachloride in mice.
