Scanning electron microscope analysis of the wall-breaking of buckwheat bee pollen and its flavonoids absorption in simulated gastrointestinal fluid

Abstract: Abstract: Low temperature polymeric ultrasound was used in the breaking wall of buckwheat bee pollen and the wall breaking process was analysis using scanning electron microscope(SEM). The bee pollen cells were splited, ruptured along the germ ditch, and broken to into pieces successively in SEM field. A more accurate calculation method for wall breaking rate in SEM field was proposed. Based on SEM observation, a ratio of wall-broken 98.33% was obtained under the best conditions for breaking the cell wall: initial temperature 0 oC, ultrasound power 400 W, the pulse duty factor was 1:2, ultrasound time 30 min, the ratio of material to liquid 1:30. Further study shows that the wall-cracking rate bears a linear relationship to the total absorptance rate of bee pollen flavone in simulated gastric and intestinal fluid (the correlation coefficient R2=0.9997) .
