Determination of phospholipids content in bell pepper by UPLC-QQQ

Abstract: Abstract: A method of determination of phospholipids in bell pepper by ultra-high performance liquid chromatography-tandem mass spectrometry (UPLC-QQQ) was established. Samples were extracted with isopropanol-methanol-water and purified by liquid extraction , and then separated on a HILIC (i.d. 2.1×100 mm,1.7 μm) system, determined by ESI-QQQ. Relative quantification was carried out with PC 28:0, LPE 14:0, LPC 14:0, PG 28:0, PA 28:0, LPS 16:0 as reference materials respectively. The extraction solvent and purification method were selected in the experiment. The method which can extract most kinds and content of phospholipids was used as the pretreatment method, and the adaptability of the method was tested. The results showed that the linearity of six phospholipids was good in the range of 10-1000 ng/mL, the limit of detection was 2-300 ng/mL, and the limit of quantitative was 120-1000 ng/mL . At the additive concentration of 125 ng, 250 ng, 375 ng, the recovery was 68.04~84.17%, and the RSD was 3.40~9.60%. The method provides a reliable platform for the determination of phospholipids of pepper cold injury based on membrane lipid metabolism.
