Study on the gene expression and enzyme activity of the Pathogenesis-related proteins in Postharvest muskmelon infected by Alternaria alternata

Abstract: In this study, Alternaria alternata was inoculated to Jiashi melon and 86-1 melon to compare the difference of disease resistance. Two melon cultivars were separately inoculated with A. alternata and subsequent storage at 7 ± 1°C (RH 85% - 90%). Afterwards, lesion size was measured, microstructure of the lesion was observed, the expression levels of CmCht1, CmCht2 and CmGlu genes were investigated by quantitative PCR, and the enzymes activities of chitinase (CHT) and β-1,3-glucanase (GLU) were measured. The results revealed that the lesions of Jiashi melon and 86-1 melon appeared on the 9th day and 6th day. The lesion size in Jiashi melon was smaller than that in 86-1 melon during the whole storage period, and the lesion size of peel was smaller than in pulp. Moreover, lesion size of peel and pulp in 86-1 melon was 1.34 times and 1.39 times greater than that in Jiashi melon on the 30th day. It was also found that the cell structure of the peel and pulp in two melon cultivars were damaged with varying degrees. The cell wall of peel was thickened to resist the infection, however, the pulp was weaker than peel. During the late period, the cell wall of peel and pulp were stratified and shrunk, and filled with hyphae and spores. Moreover, some of the pulp cells were disintegrated and were mostly unable to recognize. The relative expression levels of CHT and GLU genes in Jiashi melon were higher than 86-1 melon during the storage period, and peel was higher than pulp. The relative expression level of CmCHT1 was higher during the early to the middle period, and the relative expression level of CmCHT2 was higher during the middle period, which suggesting that infection with A. alternata stimulated the expression of CHT gene. Moreover, the relative expression levels of CmGLU was higher during the middle to the late period, which suggesting that infection with A. alternata stimulated the expression of GLU gene. After infection with A. alternata, the enzymes activities of CHT and GLU in peel and pulp tissue were significantly increased and decreased with storage time, Jiashi melon was higher than 86-1 melon, and peel was higher than pulp. All these results reveled that Jiashi melon showed stronger disease resistance than 86-1 melon and that was higher in and peel than in pulp, which was closely related with the expression levels and enzymes activities of CHT and GLU.
