Observation Study on Conchiolin Enzymolysis Effects of The Two Kinds of Plant Protease

Enzymolysis test about the conchiolin extracted from the shell nacre of pearls by using pineapple protease and papain was made to examine the UV absorbance of peptide and tyrosine at the wavelength of 275 nm with ultraviolet spectrometry.After observing and analyzing conchiolin enzymolysis effect of the two kinds of plant protease,we probed into the conditions of its enzymolysis.The conclusion showed.The two kinds of plant protease have had particular enzymolysis effects and the enzymolysis effect of pineapple protease was higher than the papain’s.It appeared a line relation ship with enzymolysis products when the substrate concentration was less than 0.2%.It was optimal when the enzyme concentration was kept under 100μg/ml and enzymolysis time about 10 hours.

 The function of Baishouwu C21 steroidal glycosides(general glycosides) in resisting oxidation damage caused by endogenous free radicals was studied with exeperimental model of violent exercise mice.Five experiments were carride out,including anti-osmosis of erythrocytes, the content of peroxidated lipids and the activity of superoxide dismutase(SOD)of the liver,the activity of catalase(CAT)of erythrocytes,and the activity of GSH-peroxidase(GSH-Px)of the blood.The results indicated that exhaustive exercise cause not only the increase of the osmosis of erythrocytes and the content of peroxidated lipids of the liver,but also the decrease of the activity of all the three inner anti-oxidation enzymes namely SOD,CAT and GSH-Px;and that general glycosides could restrain from such oxidative damage induced by exhaustive exercise.This suggested that by both of their direct scavenging capacity on oxygen free radicals and the activation of the anti-oxidation.immune system,general glycosides could enhance the anti-oxidation capability of the organ and reduce the damage of free radicals to the organ to chieve anti-ageing and health benefit effect.

The activity of tyrosinase showed some relation to production of melanin.The noncompetitive inhibition of EGb on tyrosinase was determined with L-Tryosine substrate.The Kivalue was 34mg/L.The maximum inhibition rate was 52.83%at EGb concentration of 0.16mg/ml.It was hopeful that EGb might be taken as a drug in the treatment and prevention of melanin sickness.

In the paper we mainly discussed the reology of Robinia bean gum.The result showed that the viscosity of Robinia bean gum rised with the rising of concentration.When is concentration reached to 2%,viscosity was 46.4MPa.s.Its viscosity rised with the rising of the temperature and 60℃ was the optimum temperature of heating up.The effect of both freezing-thawing and pH chang was small. It had no synergsm with xanthan gum.

Polyphenoloxidase in Pinguoli pear(Pyrus bretschneideri Rehder:Pinguoli)showed reaction with odiphenol,but not with hyroguinone,m-diphenol,and mono-phenol.Maximum PPO activities showed pH 6.5,and optimum temperature for maximum PPO activity was 30℃ with catechol as a substrate.The enzyme was relatively sensitive to most inhibiors tested with the exception of NaCl and sucrose.Potassium pyrasufite was the most effective inhibitor.

 Soluble components of Juglans regial were extracted uging 95% ethanol,ethylacetate,n-hexane one by one and their antioxiative activies were detrmined using Chemiluminescence,DPPH and TBA method.The results showed that all kinds of extracts of Juglans regial have had scavenging effects on DPPH radical.The 95% ethanol extracts showed the highest activities,where as ethylacetate extracts the second.The 95% ethanol extracts gave stronger resistance to the autoxidative system of linoleate than ethylacetate ones.On the contrary the n-hexane extracts promoted the oxidation.In the linoleate per oxidative system catalyzed by Fe2+ and Fenton reaction,the inhibiting action of 95% ethanol extracts was stronger than tea polyphenols with the same concentration.Besides,95% ethanol extracts showed strong scavenging effects on superoxidized anionic radical(O2·) Produced by system of alkaline Bipheng Triphenol,but give weak scavenging effects on hydroxy1 radical(·OH) Produced by VC-Cu-barm-H2O2 system.

 Study on the antioxidation of vegetable eggplant and“SAIJIN”,a health food,made of 14 different kind of vegetables.Method 27 Wister-rats were divided into 3 groups.They were fed with artificial semi-synthetic diet added with 6% eggplant freeze-drying powder(eggplant group)and 6%"SAIJIN"Powder ("SAIJIN" group)respectively,and control group fed with normal artificial semi-synthitic diet (pair fed).All rats were Killed after 45d.Blood,liver,kidney,brain and spleen were taken.The activities of SOD,CuZn-SOD,GSH-Px and the contents of MDA in serum and tissues were assayed.Result The activities of SOD and CuZn-SOD in rats’brain of eggplant group were significantly higher than that of control.The activity of CuZn-SOD in brain of "SAIJIN"group was higher than that of control too.However,the activity of CuZn-SOD in liver of both vegetable groups was lower than that of control.Contents of MDA in serum and liver of both vegetable groups were decreased obviously.The acivities of GSH-Px were of no difference among all groups.Conclusion Eggplant and health food "SAIJIN" could improve the activity of SOD to scavenge free radicals ,inhibit lipid peroxidation,reduce the content of malonic aldehyed and strengthen the antioxidation of body.

The nutritional components of the rye (secale cereale L.)of Luozhen NO.1,including protein,amino acids,minerals and so on,were analyzed in this paper.The results showed that the rye had a protein content of 17.1%, higher than the average wheat;the rye also had diverse amino acids,especially Lys,whose content was 60%,higher than that of the average. It indicated that the rye protein was better.The analysis of minerals revealed that the rye had more minerals,especially Fe,Mn,Mg,K,Ca,Zn,whose contents were 1348.6%,460.9%,152%,137.4%,89.4% and 45%,respectively higher than those of the average.The rye,as a material of food industry that would bentfit to the health,had a wide prospect.

The antioxidant activity and water-soluble total phenols content of 38 fruits and vegetables gathered from south of china (edible portion)were assayed by using the bleaching of β-carotene-linolic acid model assy, The correlation of activity and content was compared. It was found that ginger、purple cabbage、banana、strawberry and black olive had the higher antioxidant activity. The water -soluble total phenols content of all samples ranged from 15.37 to 339.49mg/100g.The correlation between the antioxidant activity and total phenols content of all samples was r=-0.47.After classifying on the basis of the characteristic component of the samples,the correlation between the antioxidant activity and total phenols content of the anthocyanin,chlorophyll and flavonoid was r=-0.58,-0.59and -0.81respectively.So it could the inferred that there was a high correlation between the antioxidant activity and the characteristic components of the fruits and vegetables.

 Extract the Pumpkin polysaccharide from Pumpkin and analyse its ingredients by using water extraction, alcohol deposit; use Alloxan diabetes rats as animal models, compare with the already-made traditicnal Chinese medicines Xiaoke pilland Anemarrhena extracts, carry on the animal experiment of reducing blood sugar by using pumpkin polysaccharide. The result proved the pumpkin polysaccharide has a good effect to reduce the blood sugar of alloxan diabetes rats, and the source of raw meterials is rich, and the cost of extracting is low. The development of the product has a good propects.

As ascorbic acid can rdeuce Fe(Ⅲ)-PAN-S to Fe(Ⅱ)-PAN-S.We used spectriphotmetic method to detiermine ascorbic acid of the fruits in this paper.Fe(Ⅱ)-PAN-S comple has main high peaks at 549nm and 748nm in HAc-NaAc buffer solution at pH=4.5.At 748nm,the content of ascorbic acid (in 0-100μg/25ml)is just in accordance with Beer’s law.It is very convenience and has high selection using this method,especially for the determination of ascorbic acid of the fruits.

High-performance liquid chromatography (HPLC) was used to evaluate the influence of different variables affecting the hydrolysis of flavonoids in extract of Ginkgo biloba L.(EGb).The hydrolysis recovery was estimated and four kinds of quantiative methods were also been compared.The effects of concentration of HCl in methanol,time and methanol/H2O volume ratio can be interpreed by observing the variation of quantitative results.The acid-hydrolysis conditions optimized by using the orthogonal design method and the anlysis of ranges and variances were obtained as follows:sample soluted in 1.0mol/L HCl-methanol,hydrolyzed at 80℃ for 1.0h,then analyzed by HPLC.Results show that the concentration of HCl and methanol affected the hydrolysis efficiency more markedly than time,difference standard solutions and convert factor cause less imparity to quantiative results.

Estimation of potential shelf-life of packaged pasteurized milk utilizing impedance method was studied. The results showed that there was a highly correlativity between shelf-life of ’Fengxing` pasteurized milk and logarithm of CFU per milliliter.The impedance detect time (DT)after 30℃,6 hour’s preliminary incubation added with 100μl and 200μl milk sample or after 37℃ ,6 hours’preliminary incubation added with 100 to 400μl milk sample were investigated and the regressive equation was constructed for predicting the potential shelf-life. Comparing to traditional method, Impedance method was a rapid, accurate and convenient method and the whole estimation process could finished within 11~14 hours.

New techniques of the evaluation of functional food, such as XYZ evaluation system, the relative database construction and DNA microassay techniques were extensively employed in functional food in Japan. Compared with these new developments in functional food, fundamental studies on functional factors and their active mechanisms should be thoroughly intensified. These studies will provide a solid scientific base for functional food in China. The author also suggested that standardization of assay evaluation, producing procedures and applications and legislation of market management should be established using the advanced technology and experiences on functional food, which will promote the developments and approach of Chinese health food to the international functional food.
