食品体系中丙烯酰胺的研究进展:抑制剂及其抑制机理

摘 要:丙烯酰胺自从2002年在食品中发现以来,就受到广泛的关注。同时,人们对于这个可能致癌的物质也进行了大量研究,并在丙烯酰胺的形成模型生成机理、分析和检测方法、以及其致癌性和控制方法等方面取得了很大的进展。近年来,大部分的综述专注于其致癌机理、生成机理、分析方法和控制策略,而对于其抑制剂及其抑制机理之间的关系,未见系统报道。本文结合最新的文献研究,对丙烯酰胺的抑制剂和其抑制作用机理进行系统概述,为将来寻找丙烯酰胺抑制剂和其作用机理研究提供理论参考。

Research Progress of Acrylamide in Food System: Inhibitors and Inhibition Mechanism

Abstract: Considerable public concerns about acrylamide have been generated since it was firstly found in foods in 2002. Then, a large number of efforts have been devoted to study this probable carcinogen. Great progress has been made in the development of models for acrylamide formation, understanding of its formation mechanism, analytical methods, carcinogenicity and control strategies. Most reviews published in recent years only focused on the mechanism of acrylamide carcinogenicity, the mechanism of acrylamide formation and analysis and control methods. However, rare reports regarding the relationship between inhibitors and their inhibition mechanism have been published in a systematic way. This paper systematically summarizes acrylamide inhibitors and their proposed inhibitory mechanism, which will provide a theoretical reference for future study of acrylamide inhibitors and their inhibition mechanism.

热处理在食品工业中用于延长和提高食品的质量,但也常常会带来严重的不良后果,如损失维生素、必需氨基酸或其他营养成分,产生不良的口味或异味,以及一些有害物质(如杂环胺、丙烯酰胺等) [1]。 近年来,公众对食品中有害物质(包括热处理产生)的关注越来越多,主要是由于这些有害物质可以致突变、致癌和产生细胞毒效应[1]。在这些有害物质当中,丙烯酰胺由于其在1994年被国际癌症研究机构(International Agency for Research on Cancer,IARC)定为“可能对人体有致癌作用”[2],并于2002年被瑞典研究人员发现在一些食品中含量较高而引发全球的广泛关注。许多研究已经证明,在哺乳动物体外和体内实验中,丙烯酰胺的生殖毒素表现为致突变和致癌性[3-4];此外,它也可以引起动物和人类神经系统损伤[5]。丙烯酰胺是一种无色,无味的结晶性物质,分子量小,并具有良好的水溶解性[6];它存在于许多食品中,包括薯片、薯条、烤咖啡和面包类食品(面包、脆皮面包、饼干、早餐谷物)[7]。研究表明,氨基酸天冬酰胺与羰基源类物质的美拉德反应是丙烯酰胺形成的主要反应途径[8-11];Claus等[12]在2008年时候总结了丙烯酰胺合成的其他途径。多年的研究表明,食品中丙烯酰胺的合成是一个非常复杂的过程,许多因素都可能对其有影响,如前体物质(还原糖和天冬酰胺)的浓度、反应pH值、水分含量、食物的物理状态、以及工艺参数(加热时间和温度)等[2,7,9,13-15]。基于丙烯酰胺对人类可能具有致癌性,许多策略已经被用来控制其在饮食中的水平[16-20]。本综述对食品中具有丙烯酰胺抑制剂作用的食品添加剂和配料及其作用机理进行了阐述。这些食品配料包括酸化剂、盐、亲水胶体、维生素、氨基酸和蛋白质、抗氧化剂、硫醇、酚性化合物,以及一些植物成分。由于同一种抑制剂可能通过多种途径起作用,本文以主要作用机理对丙烯酰胺抑制剂进行了分类。如VC既可以通过降低pH值,也可以通过抗氧化起到抑制丙烯酰胺生成的作用。

1 酸化剂

做为丙烯酰胺抑制剂的酸化剂通常是有机酸,典型代表是柠檬酸。Jung等[21]发现经过体积分数为0.2%的柠檬酸处理的油炸和烘烤的玉米片,其丙烯酰胺的含量分别减少82.2%和72.8%。此外还发现,薯条先在1%和2%柠檬酸溶液浸泡1h后再进行油炸,其丙烯酰胺的含量分别降低了73.1%和79.7%。这是首次发现的一种有效、简单、实用的使用酸化剂以减少油炸和烘烤食品中丙烯酰胺的方法。另一项研究[22]也证实3种有机酸可以显著减少在土豆粉模型中丙烯酰胺的形成,柠檬酸抑制效果最好(78%),其次是L-乳酸(62%)和醋酸(46%)。抑制机理如下:通过降低pH值,酸味剂可能使食品中游离的非质子化的胺(—NH2)转换成质子化的胺离子
(—NH3+),从而在第一步就阻止了美拉德反应中丙烯酰胺的形成。究其原因,丙烯酰胺是从Schiffs碱转化而来,而Schiffs碱的形成是通过天冬酰胺的α-氨基部分与二羰基化合物的羰基碳的亲核加成而成。酸化剂使游离的非质子化的胺(—NH2)转换成质子化的胺(—NH3+),从而阻断亲核攻击和Schiffs碱的形成(图1)[21]。然而,这种抑制机理目前仍处于猜测阶段,缺乏足够的证据。

2 盐 类

人们考察了许多盐类对丙烯酰胺的作用,但得到的结果因食品模型不同等因素存在差异。其中,NaCl、MgCl2和CaCl2研究得最多。

据报道[22],薯片在热烫之前浸泡于1%的食盐溶液中,可以使丙烯酰胺的含量降低62%。Pedreschi[22]、Kolek[23]等推测,NaCl抑制丙烯酰胺形成是受处理过程和NaCl两方面影响的综合结果。然而,Mestdagh等[24]发现,在他们所研究模型中,食盐并不能显著减少丙烯酰胺的生成。这种矛盾的结果表明NaCl的抑制作用需要更多和更深入的研究。此外,酸式焦磷酸钠可以降低25%丙烯酰胺[24];50mmol/L亚硫酸钠可以降低成熟橄榄中85%的丙烯酰胺含量[25],并可抑制饼干中丙烯酰胺的生成[26];100mmol/L磷酸二氢钠和四硼酸钠可以使丙烯酰胺含量分别降低27%和15%[25]

CaCl2在一些模型中也可以显著降低丙烯酰胺的合成[24,27]。Mestdagh等[24]研究表明,CaCl2在模型系统中与对照组相比较(相同pH值)可以降低70%丙烯酰胺的合成;另外,在加热之前,往CaCl2中加入NaOH,仍然可以观察到50%抑制率。Ou等[28]在模拟实验和薯片模型实验中也获得相似结果;而Barbara等[29]则在模拟实验中获得类似结果。除此之外,Gökmen等[27]研究表明,在煎炸之前把薯条浸入CaCl2溶液中,丙烯酰胺的合成减少了95%,且对炸薯条的外观(颜色和质地)没有太大影响。CaCl2抑制丙烯酰胺生成的机理可能是通过降低体系pH值和阻碍二价金属离子与天冬酰胺结合而阻止Schiff碱的合成[24,27]。此外,L-乳酸钙在50μmol/g浓度时也可以显著降低丙烯酰胺的形成[24]。然而对于钙盐的作用,Casado等[25]却得到了相反的结果。他们发现50mmol/L CaCl2溶液可以显著提高橄榄汁中丙烯酰胺的含量(24%),其原因至今未明。

MgCl2是一个凝聚剂,常常用于水果和蔬菜加工[30]。Mestdagh[24]、Barbara[29]等都发现MgCl2在模拟体系和饼干中可以抑制丙烯酰胺的形成,但其抑制能力较CaCl2弱。此外,Mestdagh等[24]认为其抑制作用机理类似于CaCl2

3 亲水胶体

Zeng等[31]报道了质量分数为2%的果胶和褐藻酸、黄原胶(20%左右)在模型试验中可以显著降低丙烯酰胺的形成(>50%)。此外,大部分的亲水胶体,当它们质量分数达到5%,对于油炸小吃模型中的丙烯酰胺都有约30%的抑制率。某些丙烯酰胺抑制剂(褐藻酸和果胶)的浸泡时间对丙烯酰胺形成的影响要远远大于其浓度。如浸泡在体积分数1%褐藻酸溶液中5h的土豆条,其丙烯酰胺的含量比浸泡在5%褐藻酸溶液中1h的土豆条低1倍;对于在褐藻酸溶液中浸泡时间相同,而浸泡浓度不同的土豆条,其丙烯酰胺的含量无显著性差异。因此,浸泡时间对于褐藻酸和果胶来说,是影响油炸土豆产品中丙烯酰胺合成的一个非常重要的因素。这是第一个关于亲水性胶体用于抑制丙烯酰胺的形成的报道。

4 维生素

维生素广泛存在于食物中,在促进和维持人体健康起着非常重要的作用。Zeng等[32]研究发现,生物素(VB7)、吡哆醇(pyridoxine,PN,也称VB6)、吡哆胺(pyridoxamine,PM,也称VB6)和L-抗坏血酸(VC)对化学模拟体系的丙烯酰胺合成有潜在抑制作用(>50%)。水溶性维生素在食品模型中能够较好地抑制丙烯酰胺的生成,而脂溶性维生素抑制作用较弱;其中,B族维生素显示最好的抑制效果。这些结果表明,抗氧化作用可能不是这些维生素抑制丙烯酰胺生成的主要因素。在化学模拟体系中,PN和烟酸(VB3)是最好的两个抑制剂,其抑制率均超过70%;而在食品体系中,VB1、VB3、VB7、VC、PM和PN都能降低大约40%丙烯酰胺的生成。VB3和PN在化学模拟体系和食品体系中均能展示比较好的抑制效果,因此在食品加工中具有很大的应用前景。进一步的机理研究表明,VB3可以与丙烯酰胺前体物质或其本身反应生成它们加合物(图2),而与丙烯酰胺自身的反应为主要途径[33]。

另外一个研究也证实30μmol/L的PM、PN、吡哆醛(pyridoxal,PL)以及VC分别可以降低38%、21%、8%和27%的丙烯酰胺[34],其中PM显示最强抑制效果。比较一下这3种类型的VB6(PM、PN和PL)结构,发现它们的主要区别在于吡啶环的4位取代基,PM的4位取代基为氨甲基,PN为甲醇基,而PL为醛基(图3)。尽管仅仅在4位取代基不同,它们对丙烯酰胺的抑制作用差别却很大[34]。研究者推测PL抑制丙烯酰胺的形成是通过醛基竞争性与蛋白质的氨基在糖化位置结合而影响Schiff碱的缩合[35],同时推测PM是通过清除二羰基类化合物来抑制丙烯酰胺的生成[34]。根据Zeng等[33]的研究,我们推测其作用机理如下:3种不同的VB6与丙烯酰胺前体物质或丙烯酰胺自身相结合,形成它们加合产物,从而达到减少丙烯酰胺的作用(图4)。3种VB6的4位取代基团不同会影响它们与丙烯酰胺结合的能力:氨甲基和甲醇基属于邻对位活化基团,可以使加成反应中的碳正离子中间体稳定快速地形成;而醛基属于邻对位钝化基团,使反应中的碳正离子中间体不稳定、缓慢地形成[35]。

5 氨基酸和蛋白质

5.1 氨基磷脂和卵磷脂

Zamora等[37]报道,二棕榈酰磷脂酰乙醇胺可以同时降低天冬酰胺/葡萄糖和天冬酰胺/2,4-癸二烯醛两种不同模型的丙烯酰胺的含量;在天冬酰胺/葡萄糖模型中,0.5μmol/L的二棕榈酰磷脂酰乙醇胺可以使丙烯酰胺降低57%,而在天冬酰胺/2,4-癸二烯醛模型中,1.5μmol/L二棕榈酰磷脂酰乙醇胺则可以使丙烯酰胺降低53%。此外,豆类和蛋类卵磷脂对天冬酰胺/葡萄糖模型中的丙烯酰胺合成也有抑制作用[37],这是第一次关于氨基磷脂和卵磷脂抑制丙烯酰胺形成的研究。

5.2 游离氨基酸

Kim等[38]考察了0.5%谷氨酸、甘氨酸、L-半胱氨酸、L-赖氨酸对水溶液模拟体系和油炸的零食产品的丙烯酰胺的抑制作用,发现谷氨酸、甘氨酸、L-半胱氨酸对丙烯酰胺的抑制率分别达到95%、91%和87%,而L-赖氨酸却低于20%。原因可能是L-赖氨酸在水溶液的溶解度较小。在土豆小吃中,0.5%甘氨酸对丙烯酰胺抑制率超过70%;此外,土豆片在3%的赖氨酸或甘氨酸中浸泡1min后,再在185℃油液中炸1.5min,可降低80%的丙烯酰胺的生成[38]。以上研究结果表明,某些氨基酸可能会成为加工食品中有效的丙烯酰胺抑制剂,应用于商业小吃和薯片。氨基酸抑制丙烯酰胺的形成可能是由于它们能竞争性地消耗丙烯酰胺的前体或者通过氨基酸的亲核反应来增加消除丙烯酰胺的能力[38]。

最近,Koutsidis等[39]发现脯氨酸和色氨酸表现最强抑制活性,随后是半胱氨酸和甘氨酸,经过60min的加热处理,其抑制率分别是80%、55%和45%;他们进一步利用LC-MS对丙烯酰胺与氨基酸加合产物(amino acid-acrylamide,AA)进行检测,得到5种加合产物如下:Cys-AA,m/z 193;Pro-AA,m/z 187;Try-AA,m/z 276;Gly-AA,m/z 147;His-AA,m/z 227。此外,他们推测半胱氨酸、脯氨酸、色氨酸、组氨酸等含巯基氨基酸由于巯基的亲核性增加,显著促进它们的烷基化反应(图5),而甘氨酸在低水分的系统中减少丙烯酰胺主要是通过可用的α-二羰基化合物,特别是乙二醛,而不是其参与烷基化反应。

Zamora[40]和Liu Jie[41]等也证实丙烯酰胺可以与氨基酸发生迈克尔加成反应,前者获得2个迈克尔加成反应产物:3-(butylamino)propanamide和2-(bis(3-amino- 2-oxopropyl)amino)acetic acid,而后者得到4个产物(RP-1、RP-2、RP- 3、RP-4)(图6)。

另外,Zeng Xiaohui等[42]考察了12种氨基酸在化学模型中对丙烯酰胺的抑制作用,发现半胱氨酸、赖氨酸和精氨酸显示较好抑制作用,三者之间无显著性差异(抑制率>90%);脯氨酸的抑制率也达到50%;而其他氨基酸不能显著性抑制丙烯酰胺的合成。此外,通过对脯氨酸与丙烯酰胺结合产物的结构鉴定[42],揭示脯氨酸抑制丙烯酰胺的作用机理可能类似于VB3和VB6,即脯氨酸与丙烯酰胺前体或其自身相结合,生成它们的加合产物,其中脯氨酸与丙烯酰胺本身结合为主要途径(图7)。这在一定程度上进一步证实了前面Koutsidis等[39]的推测。然而,其他氨基酸是否都遵循这个反应机理则需要进一步的研究。

以上这些研究结果并不都吻合[24-25,38-42],说明实验设置,如加热条件、添加方式(即浸泡、热烫或搅拌均匀)等都可能影响氨基酸对丙烯酰胺形成的抑制作用[24]。

6 抗氧化剂

到目前为止,对于抗氧化剂对丙烯酰胺合成的影响,并没有统一的结论。Hedegaard等[43]发现,添加1%迷迭香水提取物、迷迭香油,或者干燥迷迭香叶子到小麦面团中,可以使馒头中的丙烯酰胺含量分别降低62%、67%和57%。他们推测迷迭香对丙烯酰胺的抑制机理可能是其含有可以捕捉羰基类化合物的物质。Ou等[44]考察了一些抗氧化剂及其氧化产物对丙烯酰胺合成的影响,包括叔丁基氢醌(tertiary butylhydroquinone,TBHQ)、丁基化羟基苯甲醚(butylated hydroxylanisole,BHA)、二丁基羟基甲苯(butylated hydroxytoluene,BHT)、阿魏酸、表没食子儿茶素没食子酸酯(epigallocatechin gallate,EGCG)、VC。实验结果[44]表明,这些抗氧化剂不能有效破坏或抑制丙烯酰胺的合成,有的甚至还有促进丙烯酰胺合成的作用;但它们的氧化产物可以通过直接破坏丙烯酰胺或其合成前体天冬酰胺而起到抑制作用。Rydberg等[45]的研究也显示,在土豆条模型中,抗氧化剂抑制丙烯酰胺的效果甚微。Becalski等[46]在5年时间里考察了加拿大地区的5个品牌的薯片和早餐麦片,发现它们的抗氧化能力与丙烯酰胺的含量没有联系。Ehling等[47]的研究也证实丙烯酰胺的合成与抗氧化剂没有明显的相关性。然而,Ciesarová等[48]却发现,丙烯酰胺的含量与所添加的香料的抗氧化能力有关。

7 硫 醇

Hidalgo等[49]分别考察了硫醇与丙烯酰胺在无氧和有氧的情况下的反应,结果表明,在无氧的条件下,硫醇(苄基硫醇和N-乙酰半胱氨酸)可以与丙烯酰胺快速反应并生成稳定的加合物;然而,在有氧的情况下,没有观察到加合产物,且丙烯酰胺的含量大幅降低。可以解释为:在有氧情况下,硫醇快速转化成相对应的硫醇自由基,它可以自身聚合也可以诱导丙烯酰胺聚合,从而使丙烯酰胺减少(图8)[49]。

8 多酚类物质

酚类化合物是一种具有至少一个芳香环的化合物,它们与一个或多个羟基取代基相连[50-51]。它们从简单的低分子量单芳环到复杂的多酚类物质,并且根据其碳原子的数目和排布、以及连接的糖和酸的数目和方式等至少可以分为16种不同的类型[51]。

8.1 黄酮类

黄酮类广泛分布于蔬菜、水果、香料以及药用植物中,并且具有多种生物活性,如抗氧化[52]、抗肿瘤[53]、抑制血小板凝集[54]、抗炎[55-56],以及酶抑制活性[57]。最近,Cheng等[58]发现番茄皮中的柚皮素(naringenin)可以很好抑制丙烯酰胺的生成,相对于对照组,要低20%~50%,并随剂量增加而增大。另一方面,柚皮素也可以降低反应体系的褐变情况[58]。进一步研究表明,柚皮素可能是通过与美拉德中间体反应而生成新的衍生物。Cheng等[58]从反应体系中分离得到2个产物,并通LC-MS/MS和NMR鉴定它们的结构为8-C-(E-propenamide)naringenin和6-C-(E-propenamide)naringenin,从而证实这种推测(图9)。总之,柚皮素作为一个弱抗氧化剂,可以有效抑制丙烯酰胺的合成,抑制途径可能是通过丙烯酰胺的合成前体或其本身。这是到目前为止对于黄酮类物质抑制丙烯酰胺比较明确的作用机理研究。

8.2 羟基酪醇(hydroxytyrosol,HT)和3,4-二羟基苯基二醇(3,4-dihydroxyphenyl glycol,DHPG)

HT和DHPG是橄榄油中两个具有强抗氧化能力的多酚类物质[59]。Casado等[25]用3种不同浓度(12、25、50mmol/L)的绿色食用橄榄汁进行丙烯酰胺抑制实验,然而,他们发现在热的橄榄汁中丙烯酰胺的含量并没有显著性变化,说明对于成熟的橄榄来说,多酚类物质可能不是有效的丙烯酰胺清除剂。

8.3 肉桂酸、棓酸、阿魏酸、香豆酸、咖啡酸、儿茶素、表儿茶素等

Bassama等[60]对肉桂酸、棓酸、阿魏酸、香豆酸、咖啡酸、儿茶素、表儿茶素等7种酚性物质在天冬氨酸-葡萄糖等摩尔体系中考察它们对丙烯酰胺合成的影响。结果表明,这些物质对体系中丙烯酰胺含量并无太大影响;而且在加热过程中,随着咖啡酸溶液抗氧化性的增强,体系中的丙烯酰胺的含量并没有减少。以上研究表明这些酚性物质不是美拉德反应产物丙烯酰胺的良好抑制剂。

9 植物提取物

关于植物提取物对丙烯酰胺的抑制作用已有一些报道。Zhang等[61]发现质量分数为0.1%和0.5%的竹叶提取物能有效降低炸鸡翅中丙烯酰胺的含量(57.8%和59.0%),且不会影响其气味、口味和颜色。类似的结论也在薯片和薯条的食物模型中得到证实[62]。Cheng等[63]考察了6种水果提取物(苹果、蓝莓、山竹、龙眼、白果肉火龙果、红果肉火龙果)对化学体系中丙烯酰胺合成的影响,结果发现苹果提取物的抑制作用最强,蓝莓、山竹、龙眼没有明显的抑制作用,而火龙果可以增加丙烯酰胺的生成。进一步成分和化学模型分析表明,苹果可能是由于含有较高的原花青素而使其抑制作用最强[63]。初榨橄榄油的邻双酚类物质含量丰富,可以很好地抑制轻度或中度油炸条件下的薯片中丙烯酰胺含量,且对薯片颜色没太大影响,因而可以作为家庭油炸用油[64]。

10 结 语

本文综述了9种类型抑制剂对丙烯酰胺合成的影响及其机理研究,尽管此类研究已经取得很大进展,仍然有许多问题亟待解决:1许多实验仅仅考察食品添加剂对丙烯酰胺的抑制作用,而对其诸如整体感官特性、营养价值、微生物安全等方面的作用却鲜有涉及。2目前用于实验室研究的食物模型,主要集中在工业加工方面,却很少有关于家庭或者餐饮行业方面的研究,而实际上家庭和餐饮行业是食品消费的主要渠道,因此在食品加工过程中很有必要对这方面进行研究。3由于分离和纯化美拉德最终产物存在难度,有关丙烯酰胺抑制机理的研究多数处于推测阶段。4对于抑制剂本身以及抑制剂与丙烯酰胺或者食品中其他物质的反应产物的安全性和毒性研究,很少受到关注,而且这方面研究多在体外进行,很少进行体内研究。
