大肠杆菌耐酸分子机制研究进展

摘 要:具有耐强酸能力的大肠杆菌在酸性胁迫条件下更易生存。这主要是由于其具有葡萄糖-阻碍耐酸系统、氨基酸依赖型耐酸系统、伴侣蛋白抗酸作用以及保持膜电荷稳定等耐酸机制。了解大肠杆菌的这些耐酸分子机制,可以为食品加工工业控制大肠杆菌的污染提供新的认识,也对一些食源性致病菌的临床预防和治疗具有积极的作用。本文就大肠杆菌耐酸分子机制的研究进行综述。

大肠杆菌(Escherichia coli)是与人们日常生活关系非常密切的一类细菌,是肠道杆菌大类中的一种,包括非致病性大肠杆菌和致病性大肠杆菌。前者是人类及动物的肠道正常菌,后者可引起食物中毒。大肠杆菌分布非常广泛,土壤表层、水体、家畜、家禽、蚊蝇等都含有致病性大肠杆菌,肉类、乳与乳制品、水产品、豆制品、蔬菜、发酵食品等各种食品都有可能被致病性大肠杆菌污染,并通过饮水和食物使人中毒 [1-4]。致病性大肠杆菌是引起食物中毒最常见的病原菌之一。

大肠杆菌有较强的耐酸性,特别是其处于稳定期的细胞能够耐受pH 2.5或更低的酸性条件数小时,王金玲等 [5]分离出了一株在pH 2.5条件下耐受29 h的O157:H7。因此,大肠杆菌很容易污染酸性或发酵食品如果汁、香肠、奶酪、酸奶、腌制食品等 [6-9];另一方面,致病性大肠杆菌能够通过哺乳动物的胃(pH 1~3)进入肠道生存繁殖而致病,给人类的健康带来威胁 [10-12]。因此,对于大肠杆菌的这种能够在强酸环境生存的机制备受关注,研究人员发现大肠杆菌存在多种耐酸机制,且多种机制及其相互间的影响构成了一个复杂的耐酸调节网络。

当大肠杆菌暴露于非致死的酸性条件下时,会诱导多种耐酸系统的作用及相关基因的表达(acid resistant systems,ARs),如葡萄糖-阻碍耐酸系统、氨基酸(谷氨酸、精氨酸、谷氨酰胺)依赖型耐酸系统,ARs可保护细胞在pH 2.0~4.0的可致死条件下存活 [11-13]。最近的研究发现大肠杆菌周质空间存在分子伴侣HdeA和HdeB,在酸性条件下,HdeA和HdeB可与蛋白质结合并防止其聚集沉淀 [14-16]。大肠杆菌的其他耐酸机制包括通过改变细胞膜成分和保持膜电荷稳定以及交叉保护的机制也对其耐酸能力起重要的辅助作用。大肠杆菌在不含氨基酸的培养基(如只含有葡萄糖的基本培养基)条件下的抗酸能力明显低于其在富含氨基酸的培养基(如Luria broth,LB)中的抗酸能力 [10,17]。氨基酸依赖型耐酸系统在富含氨基酸的培养基里能发挥有效的耐酸性,而且氨基酸依赖型耐酸系统消耗质子的能力更强。但是,若大肠杆菌生存在成分复杂的酸性食品或者哺乳动物的胃中,其多种耐酸机制将同时发挥作用。

1 葡萄糖-阻碍耐酸系统(AR1)

葡萄糖-阻碍耐酸系统(AR1)是研究人员根据当大肠杆菌在pH 5.5的基本培养基生长到稳定期(stationary phase)时,pH值迅速调整到2.5后大肠杆菌仍然能够存活的现象发现的。选择性信号因子σ s和总调控蛋白CRP(cAMP receptor protein)是抵抗酸性环境所需要的 [17-18]。此外,传递质子的F 0F 1-ATP酶(F 0/F 1protontranslocating ATPase)也参与其间,但AR1是否由F 0F 1-ATP酶系统提供能量还是未知的 [17-22]。谷氨酸和谷氨酰胺能激活此耐酸系统,葡萄糖会对其产生抑制作用;pH 8.0时,细胞会产生抑制剂阻止该系统的活化 [21]。选择性信号因子σ s由RpoS编码,RpoS是细菌一般胁迫反应的主要调控因子。可以诱导RpoS表达包括胁迫条件(如碳源和氮源饥饿、渗透压升高、低pH值、温度升高等)以及一些细胞内信号。

AR1的结构组分和保护机制仍不明确,研究人员推出其作用机制,如图1所示,一方面,酸性条件可以直接作用RpoS,诱导RpoS的翻译、抑制RpoS的降解,从而增加RpoS量的积累;另一方面,酸性条件可以抑制cAMP的表达,活化诸如PhoP/PhoQ、ArcB/RssB等双组分系统,cAMP的减少将会激活RpoS的表达,活化的PhoP/PhoQ间接调控RpoS的转录,活化的ArcB/RssB间接抑制RpoS的降解 [23-27]。因此,在酸性条件下RpoS的量将明显提高并产生一定量的酸休克蛋白(acid shock proteins,ASPs),ASPs的作用是保护和修复大分子,能提高细胞膜的稳定性 [28-29]。RpoS是葡萄糖-阻碍耐酸系统不可缺少的调节因子,而且在其他耐酸机制中也起着一定的调控作用。研究表明,大肠杆菌在高渗、pH 5.0的条件下或进入稳定期,RpoS直接或间接调控其大约500 个基因的表达,占E. coli基因组的10% [25,28-32]

2 氨基酸依赖型耐酸系统

2.1 谷氨酸依赖型耐酸系统(AR2)

谷氨酸依赖型耐酸系统(AR2)是在谷氨酸存在的情况下,依赖于两个谷氨酸脱羧酶(GadA和GadB)的异构体和谷氨酸-γ-氨基丁酸(γ-aminobutyric acid,GABA)反转运体(GadC)共同发挥耐酸作用 [19-20,31]。如图2所示,AR2的作用机制是大肠杆菌利用谷氨酸脱羧酶使谷氨酸在脱羧过程中生成GABA并消耗细胞内的H +,达到升高细胞内pH值的目的;转运蛋白GadC将GABA转运到细胞外并交换新的底物(谷氨酸)。脱羧和转运的作用就是将酸性环境下进入细胞内的质子消耗,再将产物输出细胞,并交换新的底物,如此循环,能够阻止细胞内的pH值降低,以防止细胞致死。

谷氨酸依赖型耐酸系统是大肠杆菌最有效的耐酸系统。同功酶GadA、GadB和氨基酸转运蛋白GadC共同作用发挥着消耗入侵细胞内质子的作用,从而缓解因为环境低pH值而导致细胞内pH值降低的胁迫。研究发现,3 个关键基因gadA、gadB、gadC的缺失都会显著影响细菌在pH 2~3的存活率。同时还发现,pH 2.5时,GadA和GadB任何一种脱羧酶的存在都能保证AR2发挥耐酸作用,但是当pH 3.0时,2 种脱羧酶同时存在才能保证AR2的有效性 [21]。研究还发现,在pH 5.5的EG培养基和在pH 7.0的EG培养基大肠杆菌生长到对数后期,其gadA、gadB的表达量有显著差异,gadA和gadB在pH 5.5时的表达量显著多于在pH 7.0时的表达量 [30-34]。可见,酸性条件能够诱导着几个基因的表达,反过来其高表达又能很好的增强大肠杆菌的耐酸能力。

谷氨酸依赖型耐酸系统的耐酸能力除了与这3 个关键蛋白有直接关系外,其还受到另外11 个调节基因的影响,如图3所示。这个系统是处于一个复杂的调节网络之中。GadE是关键调节基因,位于gadA和gadB、gadC的上游,由gad盒子和GadE激活子组成 [35-36]

研究证明无论在何种成分的培养基(LB、LBG、BHI)还是在不同的生长时期下,GadE对gadA和gadB、gadC的表达都起到关键的调控作用 [7]。GadE除了自我调节表达外,还存在3 条调节途径。第一条途径需要传感激酶EvgS、调节因子EvgA以及YdeO的参与,YdeO能够直接或间接地激活GadE的转录;第二条途径需要CRP和RpoS两个调节因子GadX、GadW的参与,这条环路对细胞的耐酸性起到至关重要的作用,酸性环境会抑制cAMP的表达,但是cAMP的减少将会激活RpoS的表达,进而促进GadX的表达或GadW的降解;第三条途径称为TrmE环路,TrmE在细胞内的功能并不完全明确,只知道它会诱导tRNA的改变 [7,37-39]

2.2 精氨酸依赖型耐酸系统(AR3)

精氨酸依赖型耐酸系统(AR3)与AR2作用机制相同,只不过需要外源精氨酸。AR3在低pH值厌氧条件下被诱导,由精氨酸脱羧酶(AdiA)和反转运体(AdiC)组成 [7,38,40]。在富含精氨酸的酸性培养条件下,酸诱导AdiA和AdiC的表达,大肠杆菌利用精氨酸脱羧酶使精氨酸在脱羧过程中生成胍丁胺(agmatine)并消耗细胞内的H +,达到升高细胞内pH值的目的;脱羧产物胍丁胺被转运蛋白AdiC转运到细胞外并交换新的精氨酸,如图2。脱羧消耗细胞内的质子,转运交换新的底物,如此循环,虽然质子不断地渗透到细胞质中,但细胞质中pH值仍然保持在pH 4.7±0.1,在此范围内,不会引起大肠杆菌死亡 [40]

研究发现,adiC(yjdE)基因位于相邻的调控基因adiY和adiA基因的上游,这一点与AR2中的gadC和gadB基因的位置是一致的(图4)。gadB基因编码谷氨酸的脱羧酶GadB,它们上游基因gadC编码的GadC蛋白是谷氨酸和GABA的反向转运蛋白。而恰好AdiA又是精氨酸的脱羧酶,adiC编码的AdiC蛋白与GadC一样,是一种膜反向转运蛋白 [39-41]

2.3 谷氨酰胺依赖型耐酸系统(AR4)

谷氨酰胺依赖型耐酸系统是2013年清华大学施一公教授等 [13]发现的一个新型大肠杆菌氨基酸依赖型耐酸系统。证实其在谷氨酰胺(Gln)存在的条件下,依赖于谷氨酰胺酶(YbaS)和氨基酸反向转运蛋白(GadC)共同发挥对酸的抵抗能力 [13,30,42],见图2。YbaS和GadC可被酸性pH值激活,且只在pH值小于或等于6.0时才能适当发挥功能。通过吸收L-谷氨酰胺(Gln),大肠杆菌利用YbaS将之转化为L-谷氨酸(Glu)并释放气态氨。游离氨中和质子,维持大肠杆菌细胞体的pH值稳定。GadC则负责细胞外Gln与细胞内Glu交换。通过这一耐酸系统,确保了大肠杆菌在含有丰富的谷氨酰胺的极酸性食品中生存。

大肠杆菌有2 个谷氨酰胺酶基因ybaS和yneH,诱导它们表达谷氨酰胺酶的条件是不一样的。研究者分别构建了ybaS和yneH基因的缺陷菌株,发现YneH在接近中性条件发挥作用,而在pH≤6.0时,YbaS对AR4起无可替代的作用 [13]。另外,酶促反应的产物谷氨酸能够在同功酶GadA、GadB的作用下脱羧,形成谷氨酸脱羧作用耐酸系统。谷氨酰胺酶促反应结合谷氨酸脱羧反应能消耗2 份质子,发挥更强大的耐酸能力,有效保障大肠杆菌的在酸性环境的生存。

2.4 其他氨基酸依赖型耐酸系统

大肠杆菌除了上述3 种重要的氨基酸依赖型耐酸系统外,其他氨基酸,如赖氨酸、鸟氨酸也能够发生脱羧作用,消耗细胞内的质子,其作用机制和上述脱羧作用一致 [42]。但是赖氨酸脱羧酶、鸟氨酸脱羧酶需要较高的pH值(几乎不能在pH 2.5条件下发挥作用)。最近的研究表明赖氨酸脱羧作用耐酸系统确实存在于大肠杆菌中,但是耐酸能力很低 [7,40]

3 伴侣蛋白发挥抗酸作用的耐酸系统

分子伴侣是细胞中一大类蛋白质,是由不相关的蛋白质组成的一个家系,它们介导其他蛋白质的正确折叠与装配,但自己不成为最后功能结构中的组分。最新的研究发现,在大肠杆菌的周质空间中存在着酸性条件下能够帮助周质蛋白复性的分子伴侣HdeA和HdeB [14-16,43]。HdeA具有α-螺旋结构和两亲性的特点,能被DNA结合蛋白H-NS(histone-like nucleoidstructuring protein)抑制 [44-47]。在pH<3.0的酸性环境中,HdeA能够阻止周质蛋白的酸诱导聚集对细胞起到毒害作用 [47-53]。在中性pH值条件下,HdeA以折叠态没有伴侣活性的二聚体形式出现,不能和蛋白质底物结合。当pH<3.0时,HdeA迅速解离成单体,单体结构部分展开具有伴侣活性 并且暴露出疏水性的表面,然后和底物蛋白结合阻止其酸诱导聚集 [52-54]。有趣的是,HdeA和底物结合时,其能量来源于外部pH值的变化,而不需要消耗细菌本身的能量。

HdeB是HdeA的类似物,与HdeA拥有类似的结构和功能 [41,55]。它们的区别在于:HdeA在pH 2.0左右的时候发挥重要的抗周质蛋白聚集的作用,在pH 3.0左右时二者都发挥抗周质蛋白聚集的作用,但HdeB比HdeA更加有效 [46]。HdeA和HdeB抗酸性存在差异的原因是HdeB二聚体完全解离是在pH 3.0左右时,HdeA却是在更低的pH值情况下。此外,HdeB在pH值为2.0时暴露出的疏水性表面要比HdeA暴露的程度低 [14]。HdeA和HdeB在抗酸方面存在着协同作用,实验证明hdeA、hdeB单基因的缺失都会降低细菌在酸性条件下的存活率。

4 膜成分改变及膜电荷的稳定

细胞膜是食源性病原菌抵抗环境胁迫的第一道防御屏障。食源性病原菌通过改变其膜成分来应答环境压力,使膜的流动性与其生命活动相协调 [56-57]。膜上环丙烷脂肪酸(cyclopropane fatty acids,CFA)含量的变化对于大肠杆菌的耐酸性起着重要的作用 [57-58]。研究证明,环丙烷脂肪酸的增加提高了膜的稳定性,限制了酰基链整体的移动,最终减弱了膜的流动性 [56,59]

大肠杆菌不仅通过改变其膜成分来适应酸压力,而且能在一定程度上保持细胞膜电荷的稳定,避免了细胞的超极化现象(正常情况下,大肠杆菌的膜电势为负) [11,60]。大肠杆菌细胞膜上的氢/氯离子转运蛋白(CLC H +/Cl )对氨基酸依赖型耐酸系统起着重要的辅助作用,其作用的机制是:一方面,脱羧反应和酶促反应能够消耗细胞膜内的质子,降低电荷;另一方面,氢/氯离子转运蛋白能够将细胞内的氢离子转运出去,并将细胞外与氯离子类似的阴离子转运进入细胞内,中和电荷,最终使细胞内的电荷恢复到正常的负电荷 [60-63]

5 交叉保护

在一种压力下可以产生抵抗其他压力的能力,称为交叉保护,大肠杆菌存在多种交叉保护的现象(表1)。经过酸适应的大肠杆菌,可以增强抵抗其他逆境的能力 [64-65];同样地,经过诸如热、饥饿、渗透压应激的大肠杆菌,可以增强抵抗酸的能力 [66-69]。Arnold等 [67]研究证明,25 ℃饥饿培养的大肠杆菌O157:H7的抗酸能力得到提高,饥饿条件下产生的饥饿休克蛋白(staration proteins)能修复因酸而变性的大分子物质。大多数细菌之所以能发生交叉保护,原因在于细菌响应一种胁迫时会产生多种保护蛋白,保护蛋白能修复因不同物理、化学因子而变性的大分子物质 [70]

利用交叉保护这一特性,大肠杆菌能够适应多种环境的压力,而使食品防腐剂(食品防腐剂就是人为制造一种胁迫因子,而使微生物不能正常生存)失去应有的作用,增加食品中毒的现象。因此,研究不同压力之间的交叉保护,对于生产新型食品防腐剂具有重要的指导意义。

6 结 语

大肠杆菌在适应各种环境压力下,进化出了多种应激响应机制来回应各种压力信号。其中研究最多的是大肠杆菌在酸性环境的应答,它使大肠杆菌具有很强的耐受酸性胁迫能力和独特的生理功能。在过去几十年里,国内外学者潜心地研究大肠杆菌的耐酸分子机制,已经发现多种耐酸系统及其影响因素,还发现一些耐酸基因,如rpoS、gadE、gadX、phoQ等。这些基因可能作为新型的抗菌治疗靶点或者为发展新型疫苗提供依据。

随着对大肠杆菌耐酸作用机制的深入了解,也引出很多没有解决的问题,比如,AR1作用机制是什么,一切还在推测阶段;大肠杆菌的这些耐酸系统存在于其他细菌吗;环路TrmE作用途径是怎样的,TrmE为什么对gadA/BC的表达起着双重作用;对应答过程表达的一些酶类和蛋白质还没有准确的定义、其结构完全不了解;对大肠杆菌的耐酸机制的描述还在实验室阶段,然而大肠杆菌在低pH值食品中的应答机制更加复杂;还有哪些基因参与了大肠杆菌对酸信号的应答。这些问题都需要进一步的探究。但是,随着现代技术的进步,相信在接下来的几十年里,人们将在大肠杆菌耐酸方面有更进一步的发现和突破。

Recent Progress in Molecular Mechanism of Acid Resistance in Escherichia coli

Abstract:Some strains of Escherichia coli are resistant to strong acid and can survive in acidic conditions, mainly because they have a variety of acid resistance mechanisms, including a glucose-repressed system, amino-acid-dependent system, chaperone proteins and systems for stabilizing membrane charge. Understanding the molecular mechanisms of acid resistance in E. coli can provide new insight into controlling contamination in the food processing industry, and play a positive role in clinical prevention and treatment of foodborne pathogens. In this paper, the molecular mechanisms of acid resistance in E. coli are reviewed.
