一种榆黄蘑菌丝体多糖的结构表征及其降血糖作用

Structural characterization and hypoglycemic effect of a polysaccharide from Pleurotus citrinopileatus mycelium

摘要: 为探究榆黄蘑菌丝体多糖的结构表征以及降血糖作用,本文从榆黄蘑菌丝体中分离纯化得到多糖PCP-W,解析其结构;建立Ⅱ型糖尿病模型小鼠,灌胃PCP-W 8 w后,检测小鼠的空腹血糖水平、糖耐量和空腹胰岛素水平,计算定量胰岛素敏感性检查指数(Quantitative insulin sensitivity check index,QUICKI);检测小鼠血脂含量、氧化应激水平和肝脏己糖激酶(Hexokinase,HK)、丙酮酸激酶(Pyruvate kinase,PK)活性。结果表明,PCP-W相对分子质量为5 238 Da,由D-葡萄吡喃环构成,单糖组成为甘露糖、核糖、鼠李糖、葡萄糖醛酸、半乳糖醛酸、葡萄糖、半乳糖和岩藻糖,摩尔比0.003:0.010:0.011:0.015:0.001:0.944:0.013:0.001。PCP-W能显著降低糖尿病小鼠空腹血糖含量、改善葡萄糖耐受,升高空腹胰岛素水平,改善QUICKI(P < 0.01);PCP-W能显著改善糖尿病小鼠的血脂代谢和氧化应激水平,提高PK和HK的活性(P < 0.01)。因此,PCP-W具备开发成降糖药物的潜力。

Abstract: In this paper, a purified polysaccharide from the mycelia of Pleurotus citrinopileatus, used to be investigated its structure characterization and hypoglycemic effects, named PCP-W, was extracted by hot water method followed by purification via sephadexa-25 DEAE ion exchange and Sephacryl S-200 gel filtration chromatography sequentially. The purity and relative molecular weight, structure, as well as monosaccharide composition of PCP-W were analyzed by high performance size exclusion chromatography (HPSEC), infrared spectrum scanning and 1-phenyl-3-methyl-5-pyrazolone (PMP) precolumn derivatization, respectively. Type II diabetic mice, induced by intraperitoneal injection of Streptozotocin (STZ) combined with high-fat diet, were oral administered at low (50 mg/kg), medium (100 mg/kg) and high (200 mg/kg) doses , respectively, and fasting blood glucose content in mice was measured weekly. After 8 weeks of treatment, oral glucose tolerance test (OGTT) was performed,the fasting insulin level was detected, and quantitative insulin sensitivity check index (QUICKI) was calculated. Oral fat tolerance test (OFTT) and fasting blood lipid content were measured to evaluate the effect of PCP-W on blood lipid metabolism in diabetic mice. The antioxidant indexes of serum and liver and the activities of hepatic hexokinase (HK) and pyruvate kinase (PK) were detected. The results showed that the purity of PCP-W was 99.89% and the relative molecular weight 5238 Da, composed of D-glucopyran ring. The monosaccharides were mannose, ribose, rhamnose, glucuronic acid, galacturonic acid, glucose, galactose and fucose, with the molar ratio as following: 0.003:0.010:0.011:0.015:0.001:0.944:0.013:0.001. After 8 weeks of oral administration of PCP-W, compared with the diabetic mice, the blood glucose and the area under curve (AUC) of OGTT of mice in the low, medium and high dose groups decreased significantly, the fasting insulin level increased, and QUICKI improved markedly(P < 0.01). PCP-W could significantly reduce the AUC of OFTT (P < 0.01), triacylglycerol (TG), total cholesterol (TC), free fatty acids (FFA), low density lipoprotein cholesterol (LDL-C) (P < 0.05), and increased high density lipoprotein cholesterol (LDL-C) (P < 0.05). Meanwhile, in the mice treated by PCP-W, the content of malondialdehyde (MDA) (P < 0.01) was reduced, the activities of catalase (CAT), superoxide dismutase (SOD) and glutathione peroxidase (GSH-Px) rose, and the total antioxidant capacity (T-AOC) ameliorate was enhanced both in serum and liver. In addition, after treatment of PCP-W, the activities of hepatic PK and HK of the three dose groups were significantly increased (P < 0.01). Therefore, PCP-W may have a good hypoglycemic effect and the potential to develop hypoglycemic drugs. This study provides a theoretical basis for the further development of P. citrinopileatus.
