Quality Analysis of Flax Lecithin Based on High Performance Thin Layer Chromatography

Abstract: Purpose:To establish Quality Analysis method of HPTLC for Lecithin from flax oil foot. Methods: The lecithin sample and phosphatidylcholine standard were prepared into solution with chloroform-methanol (3:2,V:V) and chloroform:methanol:water (65:25:4,V:V:V)was used as developing system. The effect of different chromogenic agent, different developing system,different spotting sample volume, different thin layer chromatography plate,different temperature and different humidity on separation of each composition in lecithin sample were investigated.Using the best developing condition,the lecithin sample and phosphatidylcholine standard were developed, the peak area of phosphatidylcholine was scanned, standard curve was drawn and the content of phosphatidylcholine in lecithin sample was determined by external standard method. Results:The results shows that the separation of each composition in lecithin sample was suitable for qualitative anlysis, the average content of phosphatidylcholine in lecithin sample is 63.33%, with precision RSD of 1.82% and 3.73%,stability RSD of 1.60% and the average recovery of 98.5%. Conclusions: The method is rapid, accurate and reproducible, can provide new techenology reference for quality analysis of lecithin.
