Simultaneous determination of vitamins A,D and E in food for special medical purposes by online solid phase extraction combined with two-dimensional liquid chromatography

Abstract: A rapid method for the determination of vitamin A, D and E in food for special medical purposes has been established. The study was based on the online solid phase extraction combined with two-dimensional liquid chromatography. The sample solution was directly injected into the HPLC to analyse after pretreated by hot saponification, and the online solid phase extraction was for purifying and enrichment of vitamin A, D and E.The solid phase extraction column packing, the ratio of loading and eluting solvent was examined, moreover, the separation conditions of two-dimensional chromatography were systematically optimized. The results indicated that excellent linearity [correlation coefficient (R2) ≥0.997] were achieved for various vitamins quantification in the concentration range. The recoveries of the method (84.4%-109.0%) and the relative standard deviation [RSD, 0.34%-5.55% (n=6)] meet the performance criteria required for the determination. The method has the higher automation, lower detection error and better efficiency,and can be used for the analysis and detection of vitamins in special medical food.
