摘要: 以间氯过氧苯甲酸(m-chloroperbenzoic acid,MCPBA)为氧化剂,使黄曲霉毒素G1(Aflatoxin G1,AFG1)转化为其8,9-环氧化物,与牛血清白蛋白(bovine serum albumin,BSA)在两相反应体系中实现偶联,得到复合物AFG1-BSA。复合物紫外扫描结果显示其扫描图谱与BSA和黄曲霉毒素G1的图谱有明显差异,同时复合物与BSA荧光强度的差异,这表明BSA与黄曲霉毒素G1偶联。AFG1-BSA中AFG1与BSA的摩尔比为4.29:1。以合成人工抗原免疫小鼠,制备了特异性的鼠抗AFG1多克隆抗血清。用间接ELISA法测得抗体血清的最高效价可达1:16000,并测定了抗体的敏感度以及特异性。

Abstract: Peracid m-chloroperbenzoic acid as the oxidant, aflatoxin G1 was transformed into AFG1-8,9-epoxide,which was coupled to BSA in a two-phase system. The UV spectrum of the conjugate AFG1-BSA is different from BSA and Aflatoxin G1 obviously, and its fluorescence intensity is different from BSA. All these showed that the preparation of AFG1-BSA is successful. The molar ratio of AFG1 to BSA in adduct is 4.29:1. BALB/c mice were immunized with AFG1-BSA for 6 weeks, and the titer antiserum from those mice were testified by indirect ELISA, as 1:16000. The sensitivity of the antibody and its cross-reactivity with AFB1-OVA were also assayed.
