应用PCR-DGGE研究玉米储藏过程中的霉菌变化

PCR-DGGE Analysis of Molds Community Composition in Stored Grain Corn

摘要: 玉米是我国最主要的储粮作物之一,但在玉米储存过程中容易发生霉菌污染问题,造成严重的经济损失。研究储粮中霉菌污染的发生规律,构建储粮中微生物生长预测模型,从而采取相应措施减少霉菌及真菌毒素的污染,是提高我国粮食安全和食品安全的重要途径。本文利用变性梯度凝胶电泳(Denaturing Gradient Gel Electrophoresis,DGGE)研究了粮库中不同贮存时间和不同空间位置的玉米样品中霉菌群落的特点,结果表明,本研究所采样的储粮玉米中霉菌污染以青霉、曲霉和毛霉为主,储粮玉米中霉菌群落的变化与其贮存时间具有较强的相关性,而与其在粮库中的空间位置相关性较小。在贮存时间方面,贮存1年和贮存3年的储粮玉米样品中霉菌群落具有较大的差异,而贮存2年样品的霉菌群落处于过渡期状态。本文初步探究的储粮玉米中霉菌群落的时空分布特征,可以为建立准确可靠霉菌群落模型提供理论支持和数据支持。

Abstract: Corn is one of the most important stored grains in China. However, in the process of storage, corn is very common to be contaminated by molds, which could cause serious economic losses. It is an important way to improve food security and food safety in our country by studying the occurrence of molds contamination and constructing prediction model of molds in stored grain to guide the prevention of mold contamination. In this article, the Denaturing Gradient Gel Electrophoresis (DGGE) was used to research the molds community composition in stored grain corn of different years and different spatial location in the grain depot. The results revealed that Penicillium, Aspergillus and Mucor were the main molds found in the stored grain corn. The years of storage were closely related to the molds community composition in stored grain corn, while the spatial location in the grain depot showed much less relationship to it. The corn samples stored for 1 year and 3 years showed great difference in their molds community composition, while the corn samples stored for 2 years were in a period of transition. The results of this study could provide important theory and data for construction of molds prediction model in stored grain.
