Determination of Mannose in Dendrobium officinale Kimuraet Migo Beverage by HPLC

Abstract: Objective: To establish a HPLC method to determine the content of D-mannose (D-Man) in Dendrobium officinale Kimura et Migo beverage (DOB). Methods: Ethanol precipitation followed by hydrochloric acid hydrolysis and derivatization was carried out for sample preparation. Mannose was separated on an Agilent Eclipse XDB-C18 (4.6 mm × 250 mm, 5μm) using a mobile phase made up of 20 mmol/L  ammonium acetate and acetonitrile (80.5:19.5, V/V) at a flow rate of 1.0 mL/min. The detection wavelength was set at 250 nm. Results: No interference was found from Panacis quinquefolium in DOB. The linear range of Man was between 7.815μg/mL and 312.6μg/mL (r = 0.9997). The average recovery was 99.66% (RSD 1.76%, n = 6). Conclusion: This method is specific, sensitive, accurate and reproducible and may therefore be applicable for the determination of D-Man in DOB.
