Construction of NIR determination model of polymalic acid in Aureobasidium pullulans fermentation

Abstract: Abstract: An NIR model to predict polymalic acid concentrations in Aureobasidium Pullulans fermentation based on near-infared spectroscopy and partial least square algorithm was constructed in this study.The optimized model was confirmed at waveband from 5638 to 6024cm-1 and a series of pre-processing methods were applied including multiplicative scatter correlation, standard normal variation, Savitzky-Golay denoising and first derivative spectrums. Internal RMSEP and Rp of the model equal 1.492 and 0.9853,respectively, with the determination of PMLA aiming at the first overtone of C-H bond in methylene and methine groups. Furthermore,this model could predict concentration of polymalic acid and could be well-used in medium optimization and mutated strains screening according to results of external tests.
