和田玉枣中环磷酸腺苷(cAMP)的分离纯化工艺研究

Separation and Purification of Adenosine 3′,5′-Cyclic Monophosphate(cAMP) from Hetian Jade Jujube

摘要: 研究新疆和田玉枣中cAMP的分离纯化工艺。结果表明:LS-200型树脂适用于和田玉枣中cAMP的分离纯化;该树脂对和田玉枣cAMP动态吸附的最佳条件为:上样液体积为8倍树脂体积,上样液pH4,最佳上样流速1ml/min,上样液浓度30μg/ml;动态解吸工艺的最佳条件为:洗脱剂35%乙醇,洗脱剂最佳流速1ml/min,洗脱剂体积为4.8倍树脂体积;和田玉枣cAMP粗品的纯度为3.47%,其在pH3的环境中比较稳定。

Abstract: Xinjiang hetian jade Chinese jujube was used as the raw material to separate cAMP,and then it was purified with several resins. It was found that LS-200 resin is the most suitable one. The optimal technical parameters of absorbing cAMP dynamically are as follows:cAMP sample volume 8 times of LS-200 resin,its pH 4,flow rate 1 ml /min,and concentraction 30μg/ml,and those of desorbing cAMP dynamically are as follows:taking 35% ethanol as eluent,its flow rate 1 ml /min,and its amount 4.8 times of LS-200 resin. The cAMP purity of freeze-dried sample after resin purification is 3.47% and it is stable at pH 4.
