Isolation, Purification and Characterization of Water-soluble Polysaccharides from Ampelopsis grossedentata

Abstract: Polysaccharide from Ampelopsis grossedentata (AGP) was obtained after being extracted with hot water, precipitated with alcohol and removed free protein with Sevag method. AGP was isolated and purified by DEAE-Sephadex A-25 and Sephadex G-200 column chromatography, two pure polysaccharides (AGP-3 and AGP-4) were gained. The results of HPLC and Sephadex G-200 column chromatography illustrated that both AGP-3 and AGP-4 were protein-bond polysaccharids with comparatively homogenous molecular weights. Also, the two polysaccharids were colored by coomassie brilliant blue G-250 and carbazole reaction.
