Mutation of Rhodotorula Induced by Ultra High-pressure and Its Fermentation Conditions for Carotene Production

Abstract: In order to improve the ability of carotene biosynthesis,this study utilized ultra high-pressure(UHP)technology to treat Rhodotorula glutinis NR06 strain.A mutant NR06-H39 was obtained by treatment at 300 MPa for 10 min.The carotene production of this mutant reached 9.64 mg/L,an increase of 53.02% compared with 6.30 mg/L of parent strain,and its quality of genetics was proved stabile by experiments.Under the optimized fermentation conditions(glucose of 40 g/L,(NH4)2SO4 of 5 g/L,yeast extract of 5 g/L,media volume of 40 ml/250 ml shake flask,inoculation size of 8%,original pH of 6.5,peanut oil of 0.4 ml/L,and tomato extract of 3 ml/L),the β-carotene production could reach 12.42 mg/L,increasing by 22.84% compared with 9.64 mg/L of the control.
