Biotransformation and Purification of Lutein

Abstract: In this research, biotransformation technology of lutein esters was investigated. Transformation effect of lutein was discussed according to various factors such as the type of culture medium, original strains, shock culture or quiescent culture, pH value and inoculation amount. Qualitative and quantitative determination of free lutein was performance by dual-wavelength TLC scanning method. The results showed that the optimal parameters were: inoculating original strains C (Penicillium) 6 rings in the revulsive culture medium at pH 3, shocking culture 15 days, and the content of free lutein could reach 56.70% in biotransformation product from 0.25% in the material. Then biotransformation product could be purified by silica gel column chromatography. The appropriate parameters were obtained as follow: 0.5g sample each time, V (petroleum ether):V(ethyl acetate)=3:2 was used as the eluting solvent at the eluent rate of 3 ml/min. Then the content of free lutein can reach 90.20%.
