Determination of Soluble Protein and Analysis of Amino Acid Compositions of Selenium(Se)-accumulated Pleurotus ostreatus

Abstract: Objective:To study content of protein in Se-accumulated Pleurotus ostreatus that added different concentrations from 10 to 50 mg/kg inorganic selenium to the culture and analyse component of amino acid in the 10 mg/kg Se-treated Pleurotus ostreatus that had content of 7 to 10 μg/g selenium/g dry body and could be sold directly and eaten safely. Method:Coomassie brilliant blue G-250 was used to determine content of soluble protein,Kjeltec Protein Analyzer was used to determine content of total protein,and amino acid auto analyzer was used to analyse amino acid composition. Results:The content of soluble protein in the attained powder of Se-accumulated Pleurotus ostreatus is evidently higher than that of the control,reaching 2.25 times; The powder of the 10 mg/kg selenium treated Pleurotus ostreatus has 7 human essential amino acids(except trptophane) accounting for about 47.44% of total amino acid,nonessential amino acid accounting for 14.39% of total amino acid and branched-chain amino acid accouting for 20.37% of total amino acid. Conclusion:Se-accumulated Pleurotus ostreatus is a functional product which has high health protection and medical utilization value.
