Conditions Optimization of Tetramethylpyrazine Producing from Bacillus Subtilis S0507

Abstract: Response Surface Method (RSM) was applied to optimize the culture conditions of Tetramethylpyrazine producing from Bacillus Subtilis S0507. After the influences of single factors on culturing conditions were investigated, the effects of culturing time, temperature and moisture content on TTMP yield at a three-variable, three-level experiments using Box-Behnken design (BBD) were observed. The BBD experiment results were analyzed using Design-Expert v8.0.1.6 software. The results indicated that the obtained optimal culturing conditions were as follows: culturing time 33.84 hour, culturing temperature 41.75 ℃ and moisture content 59.63%. Under the optimum culturing conditions, the content of TTMP was 332.70 mg/kg. The relation error between the experimental value and the theoretical value was 0.83%. It was proved to be feasible that conditions of TTMP producing form Bacillus Subtilis S0507 was optimized by response surface method.
