Macroporous Resin Purification of Saponins from the sea cucumber Pearsonothria graeffei

Abstract: The adsorption and desorption rates of 7 types of macroporous resins towards total saponins from the sea cucumber Pearsonothria graeffei were investigated. Among the studied macroporous resins, XAD761 resin was the most suitable to purify crude total flavonoid extract from Pearsonothria graeffei. When 9 BV of 2 mg/mL sample was flowed through XAD761 resin column at a speed of 0.8 mL/min, the adsorption reached dynamic equilibrium. The adsorbed saponins could be completely eluted with 5 BV of 70% (V/V) ethanol at 1 mL/min, with a desorption rate of 93.31%. After the purification, the purity of total saponins was increased from 21% to 65%.
