蛹虫草富硒条件优化及硒对其中主要活性成分的影响

Optimization of Selenium Enrichment of Cordyceps militaris and Analysis of Effects of Selenium Enrichment on Contents of Two Major Active Components

摘要:

选择对蛹虫草富硒影响较大的四个因素:培养基、装液量、培养时间和硒浓度进行四因素三水平的正交试验,以优化蛹虫草菌丝体的培养条件和富硒条件,并采用ICP-MS 方法测定蛹虫草菌丝体中的硒含量。结果表明:对菌丝体生物量影响较大的因素分别为培养时间>硒浓度>培养基种类>装液量,对富硒率影响较大的因素依次为:硒浓度>培养基种类>培养时间>装液量。最佳富硒条件为:硒浓度10μg/ml、合成培养基、培养6d 和3/5 装液量;在最优化富硒培养条件下,菌丝体中两种主要药用活性成分:虫草多糖和虫草素,其含量分别为226.3mg/g和2.22mg/g,是非富硒菌丝体中的3.4 倍和5.8 倍,能大幅提高蛹虫草的药用价值,为富硒可促进虫草多糖和虫草素等活性成分增加的机理研究提供一定的理论依据。

Abstract:

An orthogonal array design involving 9 tests of four crucial factors including medium type, fill volume of medium in a 250 ml flask, culture time and selenium concentration at three levels for maximizing selenium enrichment in mycelia of Cordyceps militaris. The selenium content in mycelia of Cordyceps militaris was determined by ICP-MS method. The effects of the four factors on biomass of mycelia declined in the following order: culture time > selenium concentration > medium type > fill volume of medium in a 250 ml flask, and those on selenium enrichment ratio in the following order: selenium concentration > medium type > culture time > fill volume of medium in a 250 ml flask. The maximum selenium enrichment was achieved by the culture for 6 d with a defined medium containing 10 μg/ml selenium at 3/5 fill proportion in a 250 ml flask and the contents of two major medicinal components, cordycepic polysaccharide and cordycepin, in cultured mycelia of C. militaris reached 226.3 and 2.22 mg/g, which were 3.4 and 5.8 times of those in normal mycelia, indicating selenium enrichment was beneficial to the sharp increase of medicinal value C. militaris.
