两种野生乳菇中超氧化物岐化酶的研究

Extraction Study on Superoxide Dismutase from Lactarius deliciosus

摘要: 本文用不同的提取方法从松乳菇、红汁乳菇的干、鲜样品中提取超氧化物歧化酶(SuperoxideDismustase,SOD)。按四因素、四水平设计正交试验,经方差分析,选择SOD的最佳提取条件。通过同一波峰下紫外吸收值的不同对两种野生乳菇SOD含量做了比较。SOD的纯化采用硫酸铵分级分离和SephadexG-100柱层析,并经丙酮二次沉淀。鉴定了SOD的类型,发现松乳菇中含三种不同类型的SOD,Fe-SOD的含量相对较高。

Abstract: This paper studied different extracting methods to extract the superoxide dismutase(SOD) from the fresh and dry sample of Lactarius deliciosus Gray and Lactarius hatsudak Tanake respectively. The orthogonal test was designed according to four factors and four levels. Through analysis of variance, the optium conditions of extracting SOD were obtained. The two Lactarius deliciosus were compared. Purification of SOD precipitated with ammonium sulfate and Sephadex G-100 column chromatography and two times with acetone and then the type of SOD was assayed among the purification products. Through the experiment it was found that the Lactarius deliciosus Gray contains 3 different SOD, but the content of Fe-SOD is comparatively high.
