高效降亚硝酸盐乳酸菌的驯化复筛及分子鉴定

摘要: 目的 筛选出高效降亚硝酸盐乳酸菌, 为今后益生乳酸菌的开发及环境保护奠定一定的理论基础。方法 从地方特有食品及动物肠道中分离纯化出13株乳酸菌, 采用盐酸萘乙二胺法对13株乳酸菌的体外降亚硝酸盐能力进行测定, 并对其进行驯化复筛和抑菌试验, 通过生理生化及16s rDNA法对所分离的降亚硝酸盐最强菌株进行细菌学鉴定。结果 得到1株编号为JS3的乳酸菌, 该菌亚硝酸盐降解率为83.39%, 经过驯化复筛及培养条件优化,其降解率高达93.47%,菌株还具有抑菌性能。经鉴定菌株为玉米乳杆菌(Lactobacillus zeae), 将其命名为L.zeae.JS3。结论 菌株JS3具有高效降解亚硝酸盐能力, 能够成为今后益生菌制剂的优良菌种。

Abstract: Objective Screening the lactic acid bacteria strains that can efficiently degrade the nitrite,and establish theoretical basis for the development of functional dairy products and environmental protection.Method Isolated and purified 13 lactic acid bacterias from the samples such as sauerkraut, cheese , tested the ability of the target strains degrades nitrite by the method of hydrochloric acid naphthalene ethylenediamine. Using biochemistry and 16 s rDNA method to identified the highest nitrite-degrading lactic acid bacteria . Result Obtained a biodegradable nitrite strain JS3 , the strain nitrite degradation rate was 83.39%, through domestication compound screening and cultivate conditions optimization, the degradation rate increased to 93.47%, the strain can also inhibit the growth of G+ and G- bacteria. Strain was identified to be Lactobacillus zeae, named L.zeae. JS3.Conclusion The strain JS3 have efficient ability of nitrite degradation, it can be used as future good strains of probiotic preparations.
