Abstract: To establish a qualitative and quantitative method for the determination of sidenafil in health food by surface-enhanced micro-raman spectroscopy. The specificity of raman spectrum was used for the qualitative determination of sildenafil. Melamine was used as internal standard to detect the content of sildenafil. The peak height ratio of sildenafil characteristic peak and melamine characteristic has a good linear relation with the concentration of sildenafil. The linear fitting correlation coefficient was 0.9969.The qualitative detection limit was 0.05 mg/L, and the quantitative detection limit was 5 mg/L. The standard recoveries of solid and liquid samples were 85.6%~92.6%, 98.7%~105.2%, and the relative standard deviation were 4.14% and 3.25%. In addition, this method was used to detect actual samples, and the results were basically consistent with those obtained by HPLC-MS. This method can be used for the qualitative and quantitative detection of sildenafil in health food. This is a method with high accuracy, sensitivity and efficiency.
