Simultaneous Determination of 5 Phenolic Acids of Phenylpropanoid Pathway in Postharvest Wax Apple (Syzygium samarangenese [Blume] Merrill & L.M. Perry) Fruit by High Performance Liquid Chromatography

Abstract: A simple and sensitive high performance liquid chromatography method was established for simultaneous determination of 5 phenolic acids (trans-cinnamic acid, p-coumaric acid, caffeic acid, ferulic acid and sinapic acid) in the lignin synthesis of harvested wax apple. The separation was achieved using a InfinityLab Poroahell C18 chromatographic column under the gradient elution with the mobile phases of methanol and acetic acid aqueous solution (pH = 2.4) at a flow rate of 0.8 ml/min and a column temperature of 25°C. The detection wavelength of UV detector was set at 279 nm and 320 nm. The method has wide linear with correlation coefficients were all above 0.9994. The detection limits were in the range of 0.01-0.03 mg/kg and the quantitate on limit were in the range of 0.03-0.09 mg/kg. The spiked recoveries ranged from 86.18%-99.05%, with RSDs less than 5%. This method can be applied to determine the 5 phenolic acids of the lignin synthesis in wax apple, which was high accuracy, sensitivity and convenience. The measured content ranges of trans-cinnamic acid, p-coumaric acid, caffeic acid, ferulic acid and sinapic acid during storage were 11.46-24.69 mg/kg、0.92-1.45 mg/kg、 2.31-5.32 mg/kg、8.86-20.58 mg/kg and 9.48-22.62 mg/kg in wax apple fruit.
