Structural Elucidation of Flavonoids in the Supercritical CO2 Extract of Black Goji Fruit by Using Ultra-high Performance Liquid Chromatography Coupled with Tandem Mass Spectrometry

Abstract: The flavonoid components of the supercritical CO2 extract of black Goji berries from Qaidam, Qinghai were analyzed by ultra-high performance liquid chromatography coupled with tandem mass spectrometry (UPLC-MS/MS) employing PeakView workstation, and their structures were determined by analysis of the tandem mass spectra, ultraviolet (UV) spectroscopic analysis and comparison of the mass spectral data with the reference standards. As a result, a total of 13 flavonoids were identified, including 7 flavonol aglycones and flavonol glycosides, 3 dihydroflavonol aglycones, 2 flavonoid aglycones and 1 dihydrogen, namely rutin, quercetin-3-β-glucuronide, hyperoside, quercetin-3-O-galactopyranoside, dihydroquercetin, dihydroiso rhamnosin, dihydrokaempferol, quercetin, apigenin, hesperetin, alizarin, kaempferol, isorhamnetin, and kaempferol. It is noteworthy that hesperetin and alizarin were found in black Goji for the first time.
