工程大肠杆菌利用甘油和乳糖高效合成2’-岩藻糖基乳糖

Efficient Biosynthesis of 2’-Fucosyllactose Using Glycerol and Lactose in Engineered Escherichia coli

摘要: 本实验在大肠杆菌BL21star(DE3)中构建2’-岩藻糖基乳糖(2’-fucosyllactose,2’-FL)的从头合成途径,通过CRISPR/Cas9系统敲除β-半乳糖苷酶基因lacZ和UDP-葡萄糖脂质载体转移酶基因wcaJ,探究不同来源α-1,2-岩藻糖基转移酶对2’-FL合成的影响,调控合成途径基因的转录水平,并进行工程菌发酵条件优化。结果表明,初始工程菌摇瓶发酵72 h后2’-FL产量为0.34 g/L,敲除lacZ和wcaJ基因后发酵72 h合成的2’-FL质量浓度提高到2.12 g/L。脆弱拟杆菌(Bacteroides fragilis)来源α-1,2-岩藻糖基转移酶WcfB产量提升效果最好,摇瓶发酵合成的2’-FL质量浓度达4.12 g/L。经发酵条件优化,工程菌BLW-2在28 ℃和异丙基-β-D-硫代半乳糖苷终浓度0.2 mmol/L条件下,摇瓶发酵合成的2’-FL质量浓度提高至5.01 g/L,5 L发酵罐中分批补料发酵合成的2’-FL质量浓度达31.2 g/L。

Abstract: In this study, the de novo synthesis pathway of 2’-FL was established in Escherichia coli BL21star(DE3). The β-galactosidase gene (lacZ) and the UDP-glucose lipid carrier transferase gene (wcaJ) were knocked out by the CRISPR/Cas9 system. The effects of various exogenous α-1,2-fucosyltransferases on 2’-FL synthesis were investigated. Subsequently, the transcriptional levels of genes involved in the synthesis pathway were regulated, and the fermentation conditions were optimized. The results showed that after fermentation for 72 h, a 2’-FL concentration of 0.34 g/L was obtained in shake flasks with overexpression of the de novo synthesis pathway genes in E. coli BL21star (DE3), and the concentration of 2’-FL was increased to 2.12 g/L by knockout of the lacZ and wcaJ genes. Expression of α-1,2-fucosyltransferase Wcfb from Bacteroides fragilis in shake flasks resulted in the highest concentration of 4.12 g/L. Under the optimized fermentation conditions: 28 ℃ and 0.2 mmol/L final isopropyl-β-D-thiogalactoside (IPTG), the 2’-FL concentration was increased to 5.01 and 31.2 g/L in a shake flask and in a 5 L fermenter under fed-batch condition, respectively.
