小球藻糖蛋白的分离纯化与性质测定

摘要: 小球藻经热水抽提、脱蛋白、乙醇沉淀得粗糖蛋白,粗糖蛋白经Sephadex-75层析得到糖蛋白纯品。糖蛋白纯品经Sephadex-200层析证明为单一物质。紫外可见光谱只出现280nm和195nm的单峰。SDS-聚丙烯酰胺凝胶电泳显示糖蛋白分子量为63.7kDa。气相色谱分析表明该糖蛋白单糖组成主要是葡萄糖、半乳糖、鼠李糖。β-消去反应证明糖和蛋白是O-糖肽键连接的。

Abstract:  The crude glycoprotein was extracted from Chlorella Pyrenoidosa with hot water, and precipitated by ethanol after deproteinization. A refined glycoproteins were prepared after further purification on Sephadex-75 column chromatography, and their homogeneity was examined with Sephadex-200 column chromatography. The ultraviolet spectra showed their characteris- tic absorption peak at 280.00nm and 195.00nm. The molecular weight was estimated to be about 63.7kDa on SDS-PAGE. It was proved that the main monosacchrides were glucose, galactose and xylose by gas chromatography, and O-glycosidic linkage was found by β-elimination reaction.
