油菜蜂花粉中不同分子量级分清除羟自由基的研究

摘要: 本实验以油菜蜂花粉为原料,利用碱性蛋白酶对花粉蛋白进行降解,并用不同截留相对分子质量的超滤膜和纳滤膜对油菜蜂花粉酶解液进行相对分子质量分级,最后用Fenton体系测定了酶解物的不同相对分子质量级分清除羟自由基的能力。实验结果表明,酶解蜂花粉对Fenton体系产生的羟自由基的清除能力大于未酶解的水提物,经膜分离后得到的相对分子质量200~10000D的级分抑制羟自由基的效果非常显著,尤其是相对分子质量5000~10000D的级分,说明油菜蜂花粉清除羟自由基的有效部位主要是在相对分子质量200~10000D的级分中。实验结果显示,通过酶解和膜分离能有效地提高花粉清除羟自由基的能力。

Abstract: In this study,rape bee pollen protein was hydrolyzed by alcalase,then the hydrolysates were fractionated by ultrafiltration and nanofiltration,and finally the scavenging hydroxyl radical abilities of the fractions were determined by Fenton system. The results indicated that the hydrolysates have higher scavenging hydroxyl radical activities compared with their origin. The fractions with molecular weight between 200 D and 10 kD show the highest scavenging activity,especially the fractions with 5 kD to10 kD molecular weight. These results suggested that the hydrolysis and deep fractionation of rape bee pollen protein can enhance its scavenging hydroxyl radical activity.
