Detection of Ampicillin Residues in Milk by Surface Plasmon Resonance-based Optical Biosensor Assay Method

摘要: 采用表面等离子体共振的生物光学传感器的方法,利用生物分子相互作用分析原理及传感器对折射率的高度敏感性,结合传感器金膜表面绑定抗氨苄青霉素单克隆抗体的技术,实现了牛奶中氨苄青霉素含量的定量检测。实验中利用微型表面等离子体共振传感器构建了测量系统,介绍了传感器表面的处理方法,并对氨苄青霉素浓度为0、10、50、100ng/ml的牛奶样品进行了测量,通过对测量结果进行分析,得到了不同氨苄青霉素浓度的传感器响应图,对牛奶中氨苄青霉素含量的最低检测限达到了50ng/ml。结果表明,基于表面等离子体共振的光学生物传感器在牛奶抗生素药物残留检测方面具有很好的应用前景。

Abstract: The surface plasmon resonance (SPR)-based optical biosensor assay for detecting the ampicilllin residues in milk is developed. The assay is based on the bio-molecular interaction analysis (BIA) and the high sensitivity to the refractive index of the sensor. Combining with the monoclonal antibody anti-ampicillin binding on the sensor surface, the assay could be performed based on the capability of quantitative detection for ampicillin. The analysis system was equipped wih miniature integrated SPR sensor. The method of the preparation of the sensor surface was introduced, and the assay was designed to cover the ampicillin concentration range from 0 ng/ml to 100 ng/ml. With analyzing the detecting results, the sensorgrams of milk samples with different concentrations of ampicillin were obtained. The limit of detection for ampicillin can reach 50 ng/ml. It is sure that this assay based on the SPR technology can be applied to the detection of the antibiotics residues in milk.
