功能性骨胶原多肽提取前处理的脱脂脱钙条件研究

Optimization of Process Condition for Defatting and Decalcification of Bovine Bone before Extraction of Functional Collagen Polypeptides

摘要: 将牛骨制备成骨泥为试验材料,对提取牛骨胶原多肽的前处理脱脂脱钙条件进行研究。比较石油醚、无水乙醇、正己烷3种脱脂剂的脱脂效果;在单因素试验基础上利用二次正交旋转组合设计优化脱钙条件。结果表明:脱脂工艺最佳条件为无水乙醇作为骨泥脱脂剂,骨泥与无水乙醇料液比1:20(g/mL)、150r/min振荡浸泡脱脂24h,骨泥脱脂率可达97.46%。骨泥柠檬酸脱钙工艺最佳条件为柠檬酸浓度0.43mol/L、脱钙时间16h、料液比1:21(g/mL),脱钙率可达96.53%。

Abstract: The defatting and decalcification of bovine bone paste before extraction of functional collagen ploypeptides was optimized with respect to the selection of defatting agent and decalcification conditions including citric acid concentration, soaking time and solid-to-liquid ratio. Absolute ethanol was found to be the best defatting solvent among the investigated three. Following one-factor-at-a-time experiments, a quadratic orthogonal rotation combination design was performed to optimize the decalcification conditions. The best results for the defatting and decalcification of bovine bone paste were obtained as a defatting rate of 97.46% and a decalcification rate of 96.53% by suspension in absolute ethanol at a solid-to-liquid ratio of 1:20 (g/mL) with shaking at 150 r/min and then decalcification for 16 h with 0.43 mol/L citric acid at a solid-to-liquid ratio of 1:21 (g/mL).
