猕猴桃果实后熟软化过程中己糖激酶基因的表达分析

摘要: 为解决猕猴桃果实采后快速软化问题,本研究以‘红阳’猕猴桃果实为试材,探究10 mmol/L N-乙酰-D-葡萄糖胺(NAcetylDglucosamine,NAG)对猕猴桃后熟软化过程中基本生理变化、己糖激酶(Hexokinase,HXK)活性和AcHXKs基因表达的影响。结果表明,与对照(CK)组相比,外源NAG处理可有效降低呼吸速率和乙烯释放量,延缓硬度和总酸的下降、果胶和淀粉水解,维持较低的可溶性固形物含量。与CK组相比,NAG同时抑制HXK活性和AcHXK3表达水平,进而影响糖和呼吸代谢过程。此外,AcHXK7-9基因被NAG诱导上调表达,且与硬度显著正相关,其可能作为糖信号分子介导猕猴桃后熟软化进程。综上,AcHXK3/7-9受NAG诱导差异表达,进而行使催化和调节功能,延缓猕猴桃后熟软化。本试验结果将为猕猴桃后熟软化机制提供理论依据,也为猕猴桃保鲜技术的研发提供新思路。

Abstract: To solve the problem of rapid softening of postharvest kiwifruit, ‘Hongyang’ kiwifruit was used in this work to study the effect of 10 mmol/L NAcetylDglucosamine (NAG) treatment on the physiological changes, hexokinase activity and AcHXKs gene expression. The results showed that compared with the control group (CK), exogenous NAG treatment could effectively inhibit respiration rate and ethylene production, delay the decline of firmness and total acid, delay the hydrolysis of starch and pectin, maintain low level of total soluble solid content. Compared with the CK group, NAG inhibits HXK activity and AcHXK3 expression level, thereby affecting sugar and respiratory metabolism processes. Moreover, the expression of AcHXK7-9 were upregulated, which were positively correlated with firmness, and probably involved in glucose sensor-medicate kiwifruit postharvest softening. In conclusion, NAG treatment can induce the differential expression of AcHXK3/7-9, thereafter delay kiwifruit postharvest softening via catalytic and regulatory functions. The results may provide a theoretical basis for kiwifruit postharvest softening, and also help us develop novel strategies for kiwifruit preservation.
