Separation, Purification and Amino Acid Composition Analysis of Zinc-rich Water-soluble Protein ZnP6B-G75-WPⅡfrom Water Shield Leaves

Abstract:

A zinc rich water-soluble protein fraction named ZnP6B-G75-WPⅡwas separated from a zinc-rich water-soluble protein WP Ⅱ extracted from water shield leaves by Sephadex G-75 column chromatography and purified by Sepharose 6B column chromatography. The purity and the amino acid composition of ZnP6B-G75-WPⅡwere analyzed by SDS-PAEG and RT-HPLC, respectively. SDS analysis indicated that the molecule of ZnP6B- G75-WPⅡ was composed by a single subunit. The relative content of zinc was 8.44 mg/kg. ZnP6B- G75-WP Ⅱ was composed of 12 amino acids including glycin of high content and others such as histidine, cysteine, selenium-cysteine, arginine etc. which are related with zinc combination and selenium-cysteine was found to be contained in the protein.
