Rapid Analysis of Volatile Compounds in Daqu by Headspace Solid-phase Microextraction Coupled to Gas Chromatography-mass Spectrometry

Abstract: In order to analyze the composition of volatile compounds in the Luzhou Laojiao Daqu by headspace solid phase micro-extraction coupled with gas chromatography mass spectrometry detection (HS-SPME-GC-MS), the effects of SPME fiber type, adsorption time, and extraction temperature on volatile extraction were investigated. Using a 50/30μm DVB/CAR on PDMS fiber for 30 min headspace adsorption at 50 ℃ provided optimum extraction for the Daqu. Then the fiber was introduced into the injection port of the GC-MS system (at 250 ℃ for 5 min). Fifty-five compounds were isolated and identified, among which alcohols, esters, and ketones were the prominent ones. In addition, some pyrazines were also found. In conclusion, the method is simple, fast and accurate with high reproducibility and sensitivity.
