Antioxidant Activity of Ablmoschus manihot Flower Ethanolic Extract and Its Inhibitory Effect on the Growth of Hela Cells

Abstract: Objective: To explore the antioxidant activity of Ablmoschus manihot flower ethanol extract (AFEE) and its growth inhibitory effect on HeLla cells. Methods: Fenton reaction, pyrogallol auto-oxidation and DPPH radical-scavenging systems were used for antioxidant evaluation of AFEE in vitro. The reducing power was evaluated by potassium ferricyanide assay. Growth inhibitory effect of AFEE on Hela cells was assessed by MTT assay. Results: The scavenging activity of AFE on hydroxyl, superoxide anion and DPPH free radicals increased as the total flavonoids concentration increased. The EC50 of AFEE in scavenging hydroxyl, superoxide anion and DPPH free radicals were 170, 20 mg/L and 8.7 mg/L, respectively. The reducing power of AFE was two-fold higher than that of vitamin C. The growth of HeLa cells was remarkably inhibited by AFEE in a dose-dependent manner and the IC50 of AFEE in inhibiting Hela cell growth was 228 μg/mL. Conclusion: AFEE shows strong antioxidant activity and good inhibitory effect against the growth of Hela cells, suggesting a potential to be used as a natural antioxidant agent.
