The Breeding of High-yield Pectinase Producing Baterial Using He-Ne Laser and Studies on Its Optimal Fermentation Conditions

Abstract: A mutant strain ZH-2 with high yield of pectinase was selected by He-Ne laser mutagenesis on protoplasts of the original one. The optimal fermentation conditions were investigated: initial pH7.0, temperature 33℃, with 2% citrus peel powder and 1% lactose as carbon source, 1% (NH4)2SO4 and 0.3% yeast extract paste as nitrogen source, after fermenting for 32h on a rotary shaker. By preliminary analysis of the pectinase produced by ZH-2, optimum conditions for enzyme catalysis were pH6.5 at 50℃. It was stable and even retained 90% activity after incubation at 50℃ for 60min.
