Segregation and Purification of Polysaccharides from Phellinus igniariu and Structure Appraisal of This Fraction

Abstract: In this dissertation, we studied anion and cation exchange and to purification of polysaccharides from Phellinus igniariu. The polysaccharide structural and some physicochemical characters of this fraction were researched. Based on experimental results and general consideration, it was suitable to adopt using anion and cation exchange resin in series once. After that, 94.96% protein of crude polysaccharides could be removed, the recovery rate of polysaccharide was 77.77%. Higher molecular weight (HHM) and lower molecular weight(HLM) were gained by gel chromatograph. The optical rotation degree of Phellinus Igniariu polysaccharides [α]D25=+64.8°, [α]D25=+58.4°. Finally we study the structural of polysaccharides through IR, the results showed that the main polysaccharide chain was β-pyran-polysaccharide. Besides the monose of HLM through TLC analysis are glucose and galactose. The monose of HHM through TLC analysis are glucose.
