Flavonoids Extraction from Apocynum venetum by Ultrasonic-assisted and Purification by High-speed Counter-current Chromatography

Abstract: A novel flavonoids extraction process from Apocynum venetum by ultrasonic wave was tested. The optimum extraction conditions of flavonoids were investigated with orthogonal test design. The results showed that the optimal conditions are: 300 W ultrasonic wave, 15:1 of 60% ethanol to sample weight, and extraction time 15 min. High-speed counter-current chromatography was applied to the separation of flavonoids from a crude extract of Apocynum venetum. The experiment was performed with a two-phase solvent system composed of ethylacetate -ethanol -water-CH3COOH (4:1:5:0.25, V/V). From 150 mg crude extract, the 45 mg isoquercitrin is obtained with a purity of over 98.6% as determined by high performance liquid chromatography.
