Purification and Structural Analysis of Polysaccharides from Acanthus ilicifolius

Abstract: Crude polysaccharides extracted from Acanthus ilicifolius L. by hot water were decoloured and deproteinated by D301 and trichloroacetic acid. Two polysaccharide fractions named NAP(neutral Acanthus ilicifolius L. polysaccharide) and AAPB (acid Acanthus ilicifolius L. polysaccharide B)were isolated by anion-exchange chromatography DEAE-cellulose and size-exclusion chromatography of Sepharose CL-6B. The purified NAP and AAPB were proved to be homogeneous by HPGFC and their molecular weights (MW) were 11775 and 23161 respectively. IR,GC and LC were used to explore structural characters of the polysaccharides. Analysis of neutral monosaccharide composition indicated that NAP contained high proportions of arabinose, galactose and glucose, and AAPB was mainly composed of rhamnose, arabinose and galactose. The content of uronic acid in AAPB was 51.23% and that of protein in AAPB was 4.07%, which showed that AAPB is a pectin-type heteropolysaccharide.
