MSPD Clean-up and HPLC Determination of Tetracyclines in Rabbit Muscle

Abstract: The MSPD-HPLC-UV method for simultaneous isolation and detection of four tetracyclines(oxytetracycline, tetracycline, chlortetracycline and doxycycline)in rabbit muscle was developed. Blank samples and tetracyclines fortified rabbit muscle samples were ground with C18 filler in half solid state to pack the column, drip with washing, and determine by HPLC. In the method, the mobile phase is 0.01 mol/L oxalic acid and acetonitrile (85:15) with gradient elution, the flow rate 1.0 ml/min, with the detection wavelength 365 nm, and the injection volume 50μl. The limit of quantitation is 0.05μg/g. Overall recoveries are between 68.9% and 103.1%. The RSD ranges from1.2% to 21.8%. The results showed that the method is sensitive, accurate, convenient with low examination limits, which can be used to detect simultaneously the 4 tetracyclines residues in animal products.
