Effects of Auricularia auricula Polysaccharide on Scavenging Reactive Oxygen Speciesand Protecting Mitochondria

Abstract: Aim: To investigate if Auricularia auricula polysaccharide(AAP) would have the effects of scavenging superoxideanion(O-2•), antioxidation ,protecting mitochondria from injury. Methods: Lipid peroxidation and swelling of mouse liver andbrain mitochondria were induced by Fe2+-L-Cys system in vitro. Superoxide anion was generated by reduced nicotinamide adeninedinucleotide(NADH )- N-Methylphenazonium methyl sulfate(PMS)-4-Nitrobluetetrazolium chloride(NBT)system. Productof lipid peroxidation malondialdehyde(MDA) and swelling and superoxide anion production were determined spectrophotometrically.Results:AAP could inhibit MDA production,protect mitochondria from swelling,and scavenge O-2•obviously in a dose-dependent manner. Conclusion:AAP had the effects of scavenging O2-•, antioxidation and protecting mitochondria frominjury.
