Screening of a High-Yield of γ-Ployglutamic Acid-Producing Strain by Means of Both UV Light and He-Ne Laser

Abstract: Bacillus licheniformis BCPG006 was mutagenized by means of both UV and He-Ne laser. A mutant strain named as Bacillus licheniformis BCPG078 having a high ability to produce γ-polyglutamic acid (γ-PGA), was obtained by resistance screening. The γ-PGA yield of the mutant strain was 16 g/L during 72 h of shake flask culture at 37 ℃, initial pH 7 and 200 r/min, which was 2.67-fold higher than that obtained from the original strain (6 g/L). These results suggest that the mutant strain has strong genetic stability. Meanwhile, combination of UV and He-Ne laser provides an effective method for the screening of γ-PGA-producing strains.
