Protective Effect and Mechanism of Edible Grass Jiaosu on Alcohol-Induced Injury in HepG2 Cells

Abstract: To study the protective effect of edible grass jiaosu on alcohol-induced injury in HepG2 cells, the alcoholic injury model of HepG2 cells was established, and the cell viability, alcohol dehydrogenase (ADH) activity, lactate dehydrogenase (LDH) activity, malondialdehyde (MDA) content and superoxide dismutase (SOD) activity of HepG2 cells pretreated with edible grass jiaosu were detected. The effects of alcohol and edible grass jiaosu on HepG2 cells metabolites were analyzed by gas chromatography-mass spectrometry (GC-MS), and then the differential metabolites and their related metabolic pathways were identified to explore the protection mechanism of edible grass jiaosu on alcoholic injury in HepG2 cells based on cell metabolomics. Results showed that edible grass jiaosu significantly inhibited cell death induced by alcohol, which profited from the enhancement in ADH and SOD activities and the reduction in LDH activity and MDA content. A total of 11 intracellular and 16 extracellular differential metabolites were detected by cell metabolomics, the key intracellular metabolites were L-alanine, L-norleucine, L-proline, glycine, L-threonine and L-valine, the key extracellular metabolites were arabitol, D-galactopyranose, glycerol monostearate, butanedioic acid and octadecanoic acid. Edible grass jiaosu can regulate amino acid metabolism and energy metabolism to exhibit protective mechanisms against alcoholic injury in HepG2 cells. The results provided important theoretical evidence for the application of liver protective functional food with edible grass jiaosu.
