摘要: 通过对α-淀粉酶、纤维素酶、脂肪酶降解壳聚糖产物的黏均分子量的测定,确定了这三种酶降解壳聚糖的最佳工艺条件。结果表明:α-淀粉酶降解的最佳条件为时间120min,温度45℃,酶量1400U/g;纤维素酶降解的最佳条件为时间75min,温度45℃,酶量400U/g;脂肪酶降解的最佳条件为时间90min,温度45℃,酶量800U/g。同时得到了分子量小于2000的低聚壳聚糖,为进一步研究低聚壳聚糖与金属离子络合提供了条件。

Abstract: By determining viscosity average molecular weights of degradation products of chitosan by α-amylase,cellulose and lipase,the degradation conditions of the three enzymes were optimized respectively. The results showed that the optimal conditions of α-amylase degradation are 120 min,temperature 45 ℃ and enzyme amount 1400 U/g,the optimal conditions of cellulose degradation are time 75 min,temperature 45 ℃ and enzyme amount 400 U/g,and the optimal conditions of lipase degradation are time 90 min,temperature 45 ℃ and enzyme amount 800 U/g. The molecular weight below 2000 chitosan is obtained,which can be used for the further study on complex between chitosan oligosaccharide and metal ion.
