三氯蔗糖中间体6,1’,6’-三氧三苯甲基-五乙酰基蔗糖(TRISPA)的RP-HPLC分析

RP-HPLC Analysis of 6,1’,6’-Tri-O-tritylsucrose-penta-acetate(TRISPA)—An Intermediate of Sucralose Synthesis

摘要: 建立了三氯蔗糖合成过程中的中间体6,1’,6’-三氧三苯甲基-五乙酰基蔗糖(TRISPA)的RP-HPLC检测方法:采用Waters XTerraTMRP185μm,3.9×150mm色谱柱;以乙腈:水(84:16)作流动相;流速0.6ml/min;检测波长为234nm;TRISPA在0.05~0.8mg/ml范围内线性良好,检测限为5ng(S/N=3);平均回收率为99.2%;方法相对标准偏差为0.34%。该方法操作简便、快速、准确、灵敏度高,取得了很好的效果。

Abstract: A method for analysis of 6,1’,6’-Tri-O-tritylsucrose-penta-acetate(TRISPA), an important intermediate in the process of sucralose synthesis, was developed by reversed phase high performance liquid chromatography(RP-HPLC). The column was Waters XTerraTM RP18 5μm, 3.9 ×150 mm. The mobile phase was acetonitrile: water (84:16). The flow rate was 0.6 ml/min. TRISPA was determined with UV detector at 234 nm. Good linearity is obtained within the ranges from 0.05 to 0. 8 mg/ml and the minimum limit of detection is 5 ng. The average samples recovery is 99.2% and the relative standard derivation of this method is 0.34%. The results showed that the method is simple, rapid, accurate and sensitive.
